Production of doubled haploid lines in frequencies sufficient for barley breeding programs

Abstract: Winter barley (Hordeum vulgare L.) anthers were cultured on different liquid and on starch-solidified media. The optimal embryo and callus formation with different F1-lines and the cv. 'Igri' was obtained on a liquid medium with 20% Ficoll, 20 g/l maltose and barley starch. But the influence of the growth conditions of the donor plants and the genotypical differences are still enormous. The procedure has been optimized to such an extent that it can be used economically.

“…In cereal crop somatic tissue culture, sucrose is most frequently used in culture media for somatic embryogenesis. Maltose has been shown to be the most effective sugar for increasing androgenesis and green plant regeneration in rye (Flehinghaus et al 1991), wheat (Moieni 1997) and barley (Kuhlmann and Foroughi-Wehr 1989, However, our results revealed maltose (3%) to be an inferior carbohydrate source in comparison with sucrose (3%) for somatic embryogenesis of rye.…”

“…P reliminary e xperiments in camelina using basal media compositions commonly used in androgenesis indicated that there was no response from two formulations evaluated, i.e. N6 (Chu 1978) and KFWC (Kuhlmann and Foroughi-Wehr 1989). There was minimal response from NN (Nitsch and Nitsch 1969) (data not shown).…”

“…As described in a previous paper (Stival da Silva et al, 1997) this method has been used to produce an average of 3,5 green plants/100 plated anthers. According to Foroughi-Wehr & Friedt (1984) and Kühlmann & Foroughi-Wehr (1989) this rate of doubled haploid production enables the technique to be applied in barley breeding programs.…”
