Production of Anomalous Structures in Quercus Rubra L. Callus Cultures

Abstract: Internodal stem segments of 2–4‐month‐old Quercus rubra L. seedlings were cultured on Murashige and Skoog agar medium containing NAA and BA. Structures, termed organoids, were initiated from callus on medium containing 5 mg/1 NAA and 0.1 mg/1 BA. Organoids consisted of parenchymatous cells with a distinct epidermis and a continuous vascular system. Growth occurred principally by cell division throughout the parenchymatous tissue resulting in a variety of morphological shapes. Although no organized shoot merist… Show more

“…2). Similar results were obtained by other authors working with Quercus rubra [16] and Q. robur [21]. The growth regulators BA and NAA at the multiplication stage Different levels of BA (0, 0.1, 0.5, 1 and 5mgl -~) and NAA (0, 0.01, 0.1 and 0.5mg1-1) were tested in a clone of juvenile origin (12) and another one of mature origin (M13).…”

“…Initially, efforts were focused on regeneration of plants from callus cultures. Callus formation was stimulated (Jacquiot, 1952;Seckinger, et al 1979;Srivastava and Steinhauer, 1982), however, plant propagation was not achieved. A system based on in vitro multiplication of shoots from axillary buds has been developed (Chalupa, 1979(Chalupa, , 1981(Chalupa, , 1983(Chalupa, , 1984Bellarosa, 1981;Pardos, 1981;Vieitez et al, 1985).…”

“…This is especially so in the case of the stem tip or, if present, in the branches of regenerated structures (see e.g. Wadhi and Mohan Ram, 1964;Durzan et al, 1973;Durzan, 1982Durzan, , 1984Narayanaswamy and George, 1974;Favre, 1977;Seckinger et al 1979;Price and Smith, 1979;Nakano and Maeda, 1979;Kononowicz et al, 1984;Jarrett et al, 1985;Li et al, 1985;Chee and Cantliffe. 1988;Padmaja et al, 1990).…”
