Production and characterization of monoclonal antibodies against tissue specific epitopes on ABO blood group substances in saliva

Abstract: Mouse monoclonal antibodies (P4-2F, P4-5C) against ABO blood group substances in saliva were produced by immunization with ABO blood group active-glycoprotein after ethanol precipitation from heated saliva. These antibodies bound to saliva, irrespective of the ABO blood group and secretor status. Saliva diluted at least 3.2 x 10(4)-fold could be detected by ELISA using these antibodies. Tissue and species specificity of the antibodies was tested by ELISA and counterimmunoelectrophoresis and showed that the ant… Show more

“…Some polyclonal antisera specific for semen (Tsuda et al 1985), saliva (Eckersall et al 1981), sweat (Tokiwa et al 1987) and vaginal secretions (Itoh 1990) have been developed, and a few of them are used in practice. ABH antigens of body fluids were expected to be distributed on particular proteins (Sagisaka 1989;Sagisaka et al 1989Sagisaka et al , 1990, therefore monoclonal antibodies against the specific "carrier" proteins of saliva (Kimura et al 1991a) and semen (Kimura et al 1991b; Kishida et al 1991) have been also prepared for blood grouping. We established a hybridoma clone excreting an IgGI monoclonal antibody specific for the amniotic fluid.…”

“…These mAbs recognized the core protein of salival BGS and were tissue and species specific [1], and could be employed for blood grouping of A. Kimura et al: ABO from mixed body fluids saliva from mixed body fluids [2]. We think that the sandwich method with mAb against TSE of BGS may be the universal method for ABO blood grouping of individual body fluids from a mixture of body fluids.…”

“…The ABO blood group epitope in the eluate was detected to a 104-fold dilution by ELISA and this eluate was used as the immunogen. Immunization of mice with immunogen, fusion of mice lymph nodes cells with myeloma cells (P3U1) and establishment of antibody-producing hybridomas were performed essentially as described in the previous paper [1].…”
