Probucol Prevents Diabetes-Induced Retinal Neuronal Degeneration through Upregulating Nrf2

Abstract: Diabetic retinopathy (DR) is a sight-threatening complication of diabetes. This study investigated the therapeutic effect of probucol in a mouse model of diabetic retinopathy. C57BL/6 mice were rendered diabetic through Streptozotocin (STZ) intraperitoneal injection. Mice were treated with probucol (150 mg/kg, gavage administration) or vehicle (DMSO) for 12 weeks. Optical coherence tomography (OCT), fundus photography (FP), and fundus fluorescein angiography (FFA) were conducted to evaluate retinal structure a… Show more

“…Since we aimed to address oxidative stress and hypercholesterolemia with our drug combination, Probucol was chosen as the target co-drug to explore its combinatorial effect with Metformin. Probucol is reported to be a potentially efficacious drug to treat T2D 18 and related complications such as diabetic nephropathy 19 . However, its efficacy in combination with Metformin was unknown.…”

“…It is known that probucol acts as a lipid-reducing and antioxidant agent, and it has been used in the clinical treatment and prevention of cardiovascular diseases during the past few decades. Nowadays, an increasing number of researchers are focused on the neuroprotective effect of probucol (Bueno et al, 2020; Liu et al, 2020; Mamo et al, 2018; Quispe et al, 2018). Santos and colleagues (2020) have been shown a protective effect in SH-SY5Y cells against hydroperoxide-induced cytotoxicity.…”

“…In this sense, considering specifically Mn exposure, drugs that display neuroprotection mechanisms are particularly useful. Probucol is an organic compound with antioxidant properties that has been the subject of studies in recent years demonstrating anti-inflammatory, hypolipidemic, and hypoglycemic potential (Bueno et al, 2020; Liu et al, 2020; Mooranian et al, 2018; Yamashita and Matsuzawa, 2009). Probucol prevented oxidative damage and improved cognition in experimental models of neuropathologies (Santos et al, 2020), such as Huntington’s disease (Colle et al, 2012), Alzheimer’s disease (Santos et al, 2012), and PD (Moreira et al, 2013).…”
