In this study, eight lactic acid bacteria (LAB) strains, previously isolated from traditional and gluten-free sourdoughs, and selected for their potential in improving the sensory and rheological quality of bakery products, were screened against some common spoilage agents. The anti-mould activity was tested using strains of the species Fusarium graminearum, Aspergillus flavus, Penicillium paneum and Aspergillus niger. Regarding the antibacterial activity, it was assessed against four strains of the species Escherichia coli, Campylobacter jejuni, Salmonella typhimurium and Listeria monocytogenes. Furthermore, LAB strains were evaluated for their ability to produce exopolysaccharides, which are gaining considerable attention for their functional properties and applicability in different food industrial applications. A strain-specific behaviour against the moulds was observed. In particular, F. graminearum ITEM 5356 was completely inhibited by all the LAB strains. Regarding the antibacterial activity, the strains Leuconostoc citreum UMCC 3011, Lactiplantibacillus plantarum UMCC 2996, and Pediococcus pentosaceus UMCC 3010 showed wide activity against the tested pathogens. Moreover, all the LAB strains were able to produce exopolysaccharides, which were preliminarily characterized. The assessed features of the LAB strains allow us to consider them as promising candidates for single or multiple starter cultures for food fermentation processes.

The lack of rapid and effective approaches to determine the health benefits of strains is one of the main challenges affecting the selection of probiotics from large numbers of candidates. In this study, the probiotic potential of 44 Lactiplantibacillus plantarum strains isolated from different Chinese traditional fermented foods was evaluated, including acid and bile salt resistance, adhesion ability, survival in simulated human gastrointestinal transit, antioxidant activity, bile salt hydrolase (BSH) activity, and antibacterial activity. All tested L. plantarum strains showed high antioxidant capacity, BSH activity, and antibacterial activity. Among the strains, B652, C232, D444, and E932 were identified as the best comprehensive performed strains, which were selected for whole-genome sequencing, in order to provide clear information and identify key genes responsible for functional characteristics in vitro. It demonstrated that the antioxidant activity, adhesion activity, and ability to survive in the simulated gastric environment were found to be closely correlated with antioxidant enzyme encoding genes, cell-surface protein-encoding genes, and stress response genes, respectively. The numbers of functional genes present in strains might decide their performance in probiotic profile evaluation. The outcome of the study could support the development of a novel approach for the screening and identification of probiotics.

Purpose After being considered as a neglected product, agricultural waste is nowadays considered of paramount importance. It has become a source of many chemical compounds with industrial, pharmaceutical, and food applications. This study aims to evaluate the primary phytochemical content, the antioxidant properties, and the antimicrobial activities of different extracts of saffron flower waste (SFE) against bacterial and fungal strains involved in diverse pathologies in southern Morocco. Methods Total phenolic and flavonoid contents were determined. The antioxidant potentials were assessed by DPPH, FRAP, and β-carotene assays. The antimicrobial activity against four bacteria and four fungi was also evaluated. The findings in terms of the relationships between phytochemical content and all activities were depicted by PCA analyses. Results SFEs contained large amounts of phenolic and flavonoid compounds that contribute to the significant antioxidant activities. Diethyl ether (DE), n-Butanol (n-B), and Ethyl acetate (EA) fractions respectively exhibited more DPPH scavenging capacity, FRAP reducing power, and β-carotene bleaching inhibition. DE and EA have a remarkable effect mainly against Staphylococcus aureus strain, compared to Listeria monocytogenes, Escherichia coli and Klebsiella pneumoniae. Both SFEs showed antifungal antagonism towards key fungi species involved in post-harvest mold and crop yield losses. Botrytis cinerea was more susceptible than Fusarium solani, Penicillium expansum, and Penicillium digitatum. Conclusion These results reveal new data about extracts obtained from SFE that could be a potential source of natural antioxidant and antimicrobial agents opening new possibilities for their applications in the food system as a natural preservative and a sustainable alternative to conventional ingredients.
