Probing Antigen-Antibody Interactions on Electrode Supports by the Biocatalyzed Precipitation of an Insoluble Product

Abstract: The ampli®ed sensing of an antibody by an antigen monolayer-functionalized transducer by using a biocatalyzed precipitation of an insoluble product on the transducer is addressed. Faradaic impedance spectroscopy, cyclic voltammetry and microgravimetric, quartz crystal microbalance analyses are used to probe the precipitation of the insoluble product on the transducer. A dinitrophenyl, DNP, antigen monolayer is assembled on a Au-electrode, or a Au-quartz crystal, as a sensing interface for the dinitrophenyl-ant… Show more

“…The CPE is given by Z CPE = 1/Q(jx) n , where Q is the amplitude of the CPE, x is the angular frequency and n is the exponent. When n = 1, the CPE can be considered as an ideal capacitor (Q = C d ) [17,19,[33][34][35]. There is a good correspondence in C d values as measured by EIS and CV [17,19].…”

“…Electrochemical impedance spectroscopy (EIS) is an effective method for probing the interfacial properties of modified electrodes and often used for understanding chemical transformations and processes associated with the conductive supports (Dong et al, 2001;Bardea et al, 2000;Feng et al, 2000). The semi-circle diameter in the impedance spectrum is equal to the electron-transfer resistance, R et , which controls the electron-transfer kinetics of the redox probe at the electrode interface.…”

“…Electrochemical impedance spectroscopy (EIS) is an effective method for probing the interfacial properties of modified electrodes and often used for understanding chemical transformations and processes associated with the conductive supports [38][39][40]. The typical impedance spectrum (presented in the form of the Nyquist plot) includes a semicircle portion at higher frequencies corresponding to the electron-transferlimited process and a linear part at a lower frequency range representing the diffusion-limited process.…”
