Pristane‐induced arthritis. the immunologic and genetic features of an experimental murine model of autoimmune disease

Abstract: Pristane was injected intraperitoneally into mice of several strains, inducing an inflammatory seropositive arthritis in susceptible strains. The evolving histologic features included synovial hyperplasia, periostitis, and progressive marginal erosions. Multiple serologic immune abnormalities, including rheumatoid factor and anticollagen antibodies, also developed. Genetic analysis indicated that the major histocompatibility complex (H-2), C5 hemolytic complement (Hc), Newcastle disease virus-induced interfero… Show more

“…Even though the presence of palisading histiocytes surrounding necrotic nodules could not be ascertained, similar pathology has been reported with pristane-induced arthritis, MRL-lpr͞lpr, and SKG mice but not with CIA in DBA͞1 mice (11,20,21). In contrast to the SKG mice, the D1CC mice had no skin inflammation.…”

“…Because the balance between Th1 and Th2 was not significantly different in D1CC mice, aberrant expression of MHC class II together with abnormal selection of T cells such as survival of autoreactive clones will be investigated further. For example, the hCIITA transgene also might influence other types of arthritis in mice and possibly render the arthritis in other animal models, such as SKG mouse, spontaneous (20,32,33). Moreover to create even better and more mouse models of RA, D1CC mice may be backcrossed with the DRB1*0401 or DRB1*0402, and human CD4 double-transgenic mice, which lack I-A␤ Ϫ/Ϫ on the DBA͞1 background (34,35).…”

“…For example, it has been shown that the adjuvant composition determines the development of a given antigen-specific autoimmune disease [4] and that injection of adjuvant oil only can induce arthritis In genetically susceptible individuals (rats with RTl'*' [1,5] and mice with H-2q [6]), Sueh studies further indicate that in these situations the adjuvant itself is the critical agent which triggers otherwise well controlled autoreactive cells to become arthritogenic.…”

“…The second main reason for our interest in this arthritis in male DBA/l mice is that the males of this strain are particularly susceptible to collagen-induced arthritis (7) as well as pristane-induced arthritis (6). The susceptibility to collagen-induced arthritis and pristane-induced arthritis appears to depend on multiple genes, including MHC class I1 haplotype (6,7) and sex-related genes (8).…”

“…Interestingly, the DBA/l mouse appears to be particularly susceptible to both collagen-induced arthritis (7) and oil-induced arthritis (6). This susceptibility appears to be dependent on both sex-linked genes (males being more susceptible than females [S]) and non-sex-linked genes (mainly major histocompatibility complex [MHC] class I1 [6,7]), and there are no reports of any generalized immunologic aberration in this mouse strain.…”
