Percutaneous exposure of adjuvant oil causes arthritis in DA rats

Abstract: SUMMARYThe arthritogenic properties of adjuvant oil upon percutaneous administration in DA rats was investigated. Groups of rats were administered single or repeated percutaneous applications of Freund's incomplete adjuvant (FI A) or olive oil on shaved skin with or without prior abrasion of the skin. Control rats were shaved and abrased only. A transient arthritis developed in 8/16 animals after repeated applications of FIA on abrased skin. The incidence of arthritis increased to 7/8 animals when FIA was repe… Show more

“…The peak inflammation on day 4 induced by CFA is primarily due to edema formation and cellular influx (Kleinau et al, 1994). Thus, our results were similar to those reported by Rajkapoor et al (2007) and Sumanth et al (2012).…”

“…One possible explanation for the induction of arthritis following immunisation with adjuvants is that there is an increase in the activity of APC [56], leading to the presentation to autoreactive T cells of a hitherto unrecognised or ''sequestered'' endogenous antigen. The possibility that human RA could also be initiated by exposure to environmental factors with adjuvant-like activity has been highlighted by experiments in which arthritis was found to be induced in susceptible DA rats by percutaneous exposure of adjuvant oils [57], or even a mineral oil-containing cosmetic product [58]. Proteoglycan-induced arthritis (PGIA) is a model of polyarthritis induced by the injection of human cartilage proteoglycan plus adjuvant intraperitoneally genetically susceptible mice (BALB/c and C3H).…”

“…Methotrexate, an immunosuppressive agent did not inhibit FCA induced inflammation as also reported by Brown et al (1970). The peak inflammation on day 4 induced by FCA is primarily due to oedema formation and cellular influx (Kleinau et al, 1994). Besides producing inflammation, adjuvant induced arthritis is associated with pain and motor dysfunction (De Castro Costa et al, 1981).…”
