Prevalence of Vibrio parahaemolyticus , and Vibrio vulnificus in blue crabs (Callinectes sapidus ), seawater and sediments of the Maryland Coastal Bays

Abstract: Aims: To determine the prevalence of total and pathogenic Vibrio parahaemolyticus (Vp) and V. vulnificus (Vv) in blue crabs, water and sediment from the Maryland Coastal Bays (MCBs), USA. Methods and Results: Crab, haemolymph, sediment and seawater samples were collected monthly from four sites in MCBs from February 2012 through October 2012 with environmental parameters recorded. The most-probablenumber (MPN) methodology was used to enumerate Vp and Vv with presumptive colonies and the presence of virulence m… Show more

“…A most-probable-number (MPN) estimate of Vibrio sp. numbers was done using standard MPN tables for a three-tube series in triplicate (15,37). One milliliter of each tidal water sample was diluted using 9 ml of alkaline peptone water (APW), and 10 Ϫ1 to 10 Ϫ6 dilutions were made in 9 ml of APW in triplicates.…”

“…For MPN enumeration, 10 g of each mud sample was added to 90 ml of APW, and 10 Ϫ1 to 10 Ϫ6 dilutions were made in 9 ml of APW in triplicates. The tubes were incubated overnight at 37°C (15,37).…”

“…Numerous prevalence studies and outbreaks of V. parahaemolyticus infection have been extensively reported in oysters, clams, cockles, mussels, crabs, and shrimps (Newton et al, 2014; Rodgers et al, 2014; Xu et al, 2014; Malcolm et al, 2015). In finfish species, qualitative detection of V. parahaemolyticus in the anchovies ( Engraulis spp.…”
