Preservation of Black Bengal buck semen in soybean lecithin based chemically defined extender

Abstract: This experiment was conducted with the aim to study the effect of replacing egg yolk with soybean lecithin (SL) for cryopreservation of Black Bengal buck semen. Sexually matured Black Bengal buck (n = 5) were used and the ejaculates were obtained using an artificial vagina method. The semen samples were pooled and diluted in Tris extender with 5% Glycerol containing either 15% egg yolk (control group) or SL at different concentrations (1% SL, 1.5% SL and 2% SL). The semen samples were filled in straws and cool… Show more

“…Neat semen samples were evaluated for volume, sperm cell concentration (hemocytometer method), mass activity, abnormal count (Rose Bengal staining method), individual motility, and functional membrane integrity (hypo-osmotic swelling test [HOST]) immediately after collection [4,5]. After the initial evaluation, semen ejaculates were diluted 1:5 with Tris-fructose-citric acid-egg yolk-glycerol buffer and equilibrated at refrigeration temperature for 3 h. Filling and sealing of straws were done manually in French Mini Straws during the equilibration period.…”

“…AI costs less when compared to keeping breeding buck(s) in small flocks of less than 10 goats and the farmers will have access to quality elite buck semen. AI in goat is gaining popularity for the last few years in India (Konyak et al, 2018 andKarunakaran et al, 2019). While adapting AI technology, the buck selected as semen donor should have acceptable fertility, as it influences the reproductive potential of large female population.…”
