Presence of EGF growth factor ligands and their effects on cultured rat conjunctival goblet cell proliferation

Abstract: The amount of mucin on the ocular surface is regulated by the rate of mucin synthesis, mucin secretion, and the number of goblet cells. We have previously shown that cholinergic agonists are potent stimuli of mucin secretion. In contrast, there have been no studies on the control of goblet cell proliferation. In this study we investigate the presence of the EGF family of growth factors and their receptors in rat conjunctiva and cultured rat conjunctival goblet cells as well as their effects on activation of si… Show more

“…Likewise, disruption in type 2 repeats may interfere with epidermal growth factor receptor signaling of mucin-secreting goblet cells of the ocular surface. 44,45 A marginally significant increase in genotype frequencies of both SNP2 and 3 in subjects with dry eye suggested a trend that is in line with the functions associated with the corresponding TSP1 regions. Identical frequencies of these 2 SNPs also are consistent with a perfect LD between them.…”

“…We have previously reported that p38, although activated on collagen in response to strain, does not contribute to the mitogenic response (12). In contrast, the mitogenic effects of epidermal growth factor in mucin-secreting goblet cells are accompanied by activation of ERK and p38 but not AKT (46). Furthermore, in immortalized keratinocytes and a human colon cancer cell line, ERK inhibition halted proliferation, but AKT inhibition did not (47).…”

“…36 This has also been observed with other types of receptors in cultured conjunctival goblet cells. 37,38 Of note, BzATP, which in lacrimal gland acini activates P2X 7 receptors, 17 has a limited effect on these receptors in cultured MECs. Indeed, three P2X 7 receptor inhibitors had minor effects on the BzATP-stimulated increase in [Ca 2ϩ ] i .…”
