Preparative Separation and Purification of the Total Flavonoids in Scorzonera austriaca with Macroporous Resins

Abstract: Abstract:The use of macroporous resins for the separation and purification of total flavonoids to obtain high-purity total flavonoids from Scorzonera austriaca was studied. The optimal conditions for separation and purification of total flavonoids in S. austriaca with macroporous resins were as follows: D4020 resin columns were loaded with crude flavonoid extract solution, and after reaching adsorptive saturation, the columns were eluted successively with 5 bed volumes (BV) of water, 5 BV of 5% (v/v) aqueous e… Show more

“…Thus, taking efficiency into account, 3 mg/mL, 2.0 BV/h and 80% ethanol, 2 BV/h elution rate were the appropriate adsorption and desorption conditions, respectively. When the concentration of flavonoids in the effluent reaches 1/10 of the sample volume it is called the leakage point of flavonoids [ 17 ]. The dynamic LBLF adsorption curve on the PR column was “S” ( Figure 1 C).…”

“…The adsorption of macroporous resin relies on the van der Waals gravitational force between it and the adsorbed molecules (adsorbent), and works through its huge specific surface for physical adsorption, so that organic compounds can be separated by a certain solvent elution according to the adsorption force and its molecular weight size to achieve different purposes such as separation, purification, de-hybridization, and concentration. Macroporous resin can remove proteins, flavonoids, and pigments from polysaccharide solutions [ 51 , 52 , 53 , 54 ].…”

“…By contrast, macroporous resin column chromatography (MRCC) is an efficient, low-cost, eco-friendly, and fully-developed technology that is able to meet the needs of industrial production and has been widely applied to enrich and purify the natural active substances, especially flavonoids. For instance, MRCC has been successfully applied to purify flavonoids from Scorzonera austriaca [26], Abelmoschus manihot flower [27], and Platycladus orientalis (L.) Franco [28]. The MRCC has been used for the purification of matrine from S. tonkinesis [29]; however, to our knowledge, the research on the purification of flavonoids from S. tonkinensis by MRCC has not been discovered in literature.…”
