Preparation for Birth into an O2-Rich Environment: the Antioxidant Enzymes in the Developing Rabbit Lung

Abstract: SummaryTo determine if some specific "preparation for birth" occurs in the developing lung to help assure its successful adaptation to a comparatively 02-rich world at birth, we measured the activities of the antioxidant enzymes in the developing lungs of rabbit fetuses from 10 d before parturition to several days after birth. Superoxide dismutase (SOD), catalase (CAT), and glutathione peroxidase (GP) activities showed similar maturational patterns with significant increases id activity, compared with earlier … Show more

“…Among the major thiol-disulfide redox buffers within the cell, the GSH/GSSG system is considered the most important, since the concentration of GSH within the cytosol (1-11 mM) is far higher than most other redox-active compounds (44,107). For decades, however, the intracellular levels of GSH are known to regulate redox-dependent cellular differentiation, proliferation, and apoptosis and are, therefore, of central importance to the function of the cell (4,6,39,40). Until recently, the central dogma has been that low MW nonenzymatic thiols exclusively serve to counterbalance sources of ROS and to prevent (patho)physiological processes.…”

“…Premature infants are less prepared to cope with the oxygen-rich environment of extrauterine life, as their antioxidant mechanisms are underdeveloped compared to those infants born at term. 4,5 Lipids, especially those in the cell membrane, are susceptible to the high reactivity of reactive oxygen species. Lipid peroxidation results, whereby the double bonds of unsaturated fatty acids in the cell membrane are attacked by excess free radicals and results in the formation of isoprostanes.…”

“…A long-standing hypothesis has been that developmental increases in total SOD activity are a preparatory change for the more oxygen-rich environment organisms must survive subsequent to their birth (214,229). In this theory, the development-associated increase in total SOD activity is part of a programmed adaptive mechanism to enhance the survival of postnatal organisms.…”
