Preparation and Optimization of In Situ Gel Loaded with Rosuvastatin-Ellagic Acid Nanotransfersomes to Enhance the Anti-Proliferative Activity

Abstract: The objective of this study was to develop an optimized sustained-release nanotransfersomes (NTS) based in situ gel formulation of rosuvastatin (RO) combined with ellagic acid (EA) antioxidant, to enhance cytotoxic and anti-proliferative activity against tongue carcinoma. The concentrations of lecithin, Tween 80, and d-tocopherol polyethylene glycol succinate (TPGS) were considered as independent variables. Particle size, entrapment, and stability were selected as dependent variables. The obtained formulation … Show more

“…In addition, EA is found in food products conjugated with glycoside moieties [ 40 , 41 ]. Its potent health benefits are diverse, including antiproliferative [ 42 ], antioxidative, and anti-inflammatory [ 22 ] features, which can be beneficial for treating chronic diseases. However, the effects of EA on the representative chronic skin inflammatory disease AD have not previously been investigated.…”

“…If the formulation contains the gel formation, then the content will adhere to the bottom of the bottle instead of sliding or flowing down; this occurrence is noted as “+”. The least amount of NF concentration that could persuade the gel formation was projected as CIC [ 35 ].…”

“…Novel small molecules based on isocoumarins/3-alkylidenephthalides that were derived from rosuvastatin have shown promising properties for use as anti-cancer drugs in the future [ 85 ]. Furthermore, gel-loaded rosuvastatin [ 86 ], fabrication of rosuvastatin [ 87 ], and biocompatible copolymeric micelles of rosuvastatin [ 88 ] have shown an anti-cancer effect in aggressive tongue carcinoma, hepatic cancer, and breast cancer, respectively, with improved efficacy compared to rosuvastatin alone. Pitavastatin treatment induces cell death in ovarian cancer cells in which hydroxymethylglutarate coenzyme-A reductase (HMGCR) is upregulated and TP53 is mutated, and the anti-cancer effect of pitavastatin is solely mediated by the inhibition of HMGCR [ 89 ].…”
