Preparation and Characterization of Doxycycline-Loaded Electrospun PLA/HAP Nanofibers as a Drug Delivery System

Abstract: The present study aimed to prepare nanofibers by electrospinning in the system polylactic acid-hydroxyapatite-doxycycline (PLA-HAP-Doxy) to be used as a drug delivery vehicle. Two different routes were employed for the preparation of Doxy-containing nanofibers: Immobilization on the electrospun mat’s surface and encapsulation in the fiber structure. The nanofibers obtained by Doxy encapsulation were characterized using Fourier transform infrared (FTIR) spectroscopy, thermogravimetric (TG) and differential ther… Show more

“…In both media, doxy shows sustained release over 96 h. In SBF, an increasing dissolution tendency was observed with a decreasing release rate over time. The same phenomenon was also observed during the measurement in PBS, although a lower amount of dissolved active substance was detected [ 20 ]. The in vitro antimicrobial effect of the doxy-loaded nanofibers was previously demonstrated by the authors (Andrei V., et al, [ 21 ]), using a Kirby-Bauer disk diffusion susceptibility test.…”

“…For Doxycycline, the weight loss also took place in two steps. The first one started at around 170 °C while the second step occured at around 220 °C and continued until all sample was completely degraded after 600 °C [46] . Similaly, two stages of mass loss were observed at around 150 °C and 250 °C in TGA curve of Florfenicol [47] .…”

[46] Similaly, two stages of mass loss were observed at around 150 °C and 250 °C in TGA curve of Florfenicol. [47] TGA profile of DOX-FLO@AgNPs showed a gradual mass loss between 100 °C and 500 °C and a rapid weight reduction at around 600 °C could be due to both the dehydration and the decomposition of organics compounds.…”

“…Among them, electrospinning has attracted extensive attention. Meanwhile, electrospun nanofibers have their distinctive advantages such as simulating the structure of ECM [ 26 ], which determines their crucial position in biomedical fields such as wound dressing [ 27 ], antibacterial [ 28 ], biosensor [ 29 ], drug delivery [ 30 , 31 ] and tissue engineering [ 32 , 33 , 34 ]. In addition, electrospun nanofibers can provide favorable environments for stem cells to perform significant physiological functions [ 35 , 36 ], which makes them suitable candidates for BTE scaffolds.…”
