The use of Raman spectroscopy combined with saliva is an exciting emerging spectroscopy-biofluid combination. In this review, we summarize current methods employed in such studies, in particular the collection, pretreatment, and storage of saliva, as well as measurement procedures and Raman parameters used. Given the need for sensitive detection, surface-enhanced Raman methods are also surveyed, alongside chemometric techniques. A meta-analysis of variables is compiled. We observe a wide range of approaches and conclude that standardization of methods and progress to more extensive validation Raman-saliva studies is necessary. Nevertheless, the studies show tremendous promise toward the improvement of speed, diagnostic accuracy, and portable device possibilities in applications such as healthcare, law enforcement, and forensics.

In this paper, the SERS analysis technique for extracting principal components of non-structural protein 1 from its spectra is examined. The non-structural protein 1 is found a major role in the replication process of virus of flaviviridae, the cause for many viral diseases. SERS is a technique that can provide fingerprint spectral information of even a single molecule. However, the Raman spectra from SERS complicate the feature extraction process with redundant features. Principal Component Analysis is a signal processing technique, useful for filtering for the significant features while filtering off the redundant ones with minimal loss of information. Here, PCA adopting a 3-steps approach, i.e. Eigenvalue-One-Criterion, Scree test and Cumulative Percent Variance, is used to select significant principal components of NS1 from Raman spectra. It is found that principal components of NS1 from its spectra of [900x10] from SERS is found being trimmed to [9x10] by Scree test supplemented by EOC and [2x10] by CPV, with a corresponding reduction of 99% and 99.8% from the original spectral array. However, since the spectra of biological samples in actual is noisier, selection by the former of first nine components is found appropriate. So far, SERS analysis technique for detection of salivary NS1 has yet to be reported.

Raman Spectroscopy is a mean to study the molecular structure property of solid, liquid and gases from its scattering spectrum. It offers a detailed biochemical fingerprint for identifying the unknown molecule. Since the amount of inelastic scattering is infinitesimally small relative to that of the elastic scattering, the Raman signal emitted is extremely weak, rendering limited applications. Surface Enhanced Raman Spectroscopy augments the detection sensitivity of Raman spectroscopy with nano-sensing chip as substrate, enabling amplification of Raman signal by a factor of 10 4 to 10 9 . This innovation is made possible with the integration of the latest in optical sensing technology and nanotechnology. With this improvement, SERS has shown its niche in tracing molecular structure, especially in marking abnormal biological molecules such as cancer, conjunctivitis, AIDS. The quality of spectrum is highly dependent on the substrate type and laser wavelength. Our work here examines the spectral characteristics of 3 candidate substrate types and wavelength for application in salivary based disease detection. The spectrum intensity enhancement, oxidation and fluorescence effects are also investigated. This study finds that adoption of gold or Klarite® as substrate and 785nm as wavelength are appropriate for salivary based disease detection using SERS technique. The latter will be preferred if cost effectiveness is important.
