Preischemic Administration of Nonexpanded Adipose Stromal Vascular Fraction Attenuates Acute Renal Ischemia/Reperfusion Injury and Fibrosis

Abstract: Renal ischemia/reperfusion (IR) injury is a common clinical syndrome. Cell-based therapy provides a promising option to promote renal repair after IR injury. However, several challenges still remain because of the potential risks during cell culture, low retention rate after transplantation, and unclear effect on the progression of chronic kidney disease. Stromal vascular fraction (SVF) is considered as an attractive cell source. This study demonstrated that preischemic administration of uncultured SVF could i… Show more

“…Autologous SVF was isolated from epididymal adipose tissue according to our previously published protocol . Briefly, rat adipose tissue was washed thrice with ice‐cold sterile PBS, minced into small pieces, and digested with 0.075% type I collagenase at 37°C for 30‐40 minutes with gentle shaking.…”

“…Cell tracking of SVF was carried out by detecting CM‐DiI‐labeled cells according to our previous protocol . In brief, testicular tissues retrieved at 3 and 7 days after T/D were froze, cut into 5‐μm sections, and mounted with 4′‐6‐diamidino‐2‐phenylindole.…”

“…In these etiologies, due to the high clinical rates of renal IR injury, especially in the perioperative period, such as with partial nephrectomy, kidney transplantation, and intraoperative hypotension, AKI has received more attention from scholars. Large amounts of evidence have indicated that IR‐induced AKI could cause laterenal fibrosis (Guo et al, ; Kim & Padanilam, ; Zhou et al, ), a common process for the progression of CKD to end‐stage renal disease (Nogueira, Pires, & Oliveira, ). Collagen I is a heterotrimer composed of two α1 and one α2 collagen peptide that plays an important regulatory role in the process of tissue fibrosis (Wada et al, ).…”

“…Many of these stem cell treatments involved a priori radiation and/or chemotherapy preparative regimens. In addition, the application of cellular therapies to other non-cancer indications, some of which are already licensed, include GvHD (33, 34), stroke (35), inflammatory bowel disease (36) lung disease (37), lupus (38), myocardial repair (39) and recovery from ischemia [e.g., limb (40, 41) and renal (42, 43)]. Although this list is by no means complete, information obtained from these studies can be leveraged to better understand the safety attributes of stem cells and how they could be used clinically for a radiation injury.…”
