Fascin actin-bundling protein 1 (FSCN1) is an evolutionarily conserved actinbundling protein that plays a critical role in cell migration, motility, adhesion, and cellular interactions. Although multiple clinical studies have implicated the expression of FSCN1 in laryngeal squamous cell carcinoma (LSCC) progression, the precise mechanism of FSCN1 in the process has not been clearly elucidated. To define FSCN1 function, we characterized FSCN1-interacting proteins in LSCC cells by immunoprecipitation followed by mass spectrometry (MS). After data filtering, 119 proteins with expression in both the Hep-2 and TU-177 cell samples were identified as FSCN1-interacting partners. With in-depth bioinformatics analysis, we linked FSCN1 to critical cellular processes including cell adhesion, glycolysis/gluconeogenesis, regulation of protein ubiquitination, ribosomal RNA processing, and small molecule metabolism. We discuss the interactions between FSCN1 and some of the newly validated partners. The identification of these potential partners of FSCN1 expands our knowledge of the FSCN1 interactome and provides a valuable resource for understanding the functions of this protein in LSCC progression.

Purpose: Increased preoperative serum Carcinoembryonic Antigen (CEA) and Cytokeratin-19 Fragment (CYFRA 21-1) levels are associated with a poor postoperative prognosis in early-stage Non-Small Cell Lung Cancer (NSCLC). The Tumor Marker Index (TMI), calculated as the mean of the CEA and CYFRA 21-1 levels normalized to their respective diagnostic cutoff values, is a predictor of prognosis. This study aimed to validate the prognostic value of the TMI for Overall Survival (OS) and Disease-Free Survival (DFS) in a large patient population. Methods: Five-year OS and DFS based on preoperative serum CEA and CYFRA 21-1 levels and the TMI were evaluated in 454 patients (252 men and 202 women) with Stage I NSCLC (median age, 67 [range: 35-86] years) who underwent complete surgical resection. Univariate and multivariate analyses of serum CEA and CYFRA 21-1 levels, and the TMI were performed. Results:The 5-year OS and DFS rates were significantly lower for patients with elevated serum CEA/CYFRA 21-1 levels than for patients with normal CEA/CYFRA 21-1 levels. The TMI was also markedly different, demonstrating significantly lower OS and DFS in patients with a TMI of >0.5 (P<0.001). Eighty-four patients with normal CEA and CYFRA 21-1 levels had an elevated TMI, which correlated with significantly lower OS and DFS. Multivariate analysis indicated the independent prognostic value of the TMI. Conclusion:The TMI is an independent preoperative prognostic factor in Stage I NSCLC and should be considered in the decision making for postoperative therapy.

Programmed cell death-1 (PD-1) -targeted immunotherapy has become a promising treatment paradigm for patients with advanced non-small cell lung cancer (NSCLC). Clinical responses to checkpoint inhibition therapy in NSCLC have been associated with programmed death-1 ligand 1 (PD-L1) expression. However, the association between the expression of PD-L1 and PD-L2 and the clinicopathological features and patient outcomes in NSCLC remain unclear. We retrospectively analyzed 364 patients (158 squamous cell carcinoma and 206 adenocarcinoma) who underwent complete resection between 2009 and 2012. Expression of PD-L1 and PD-L2 was determined by immunohistochemistry. Correlations between PD-L1/PD-L2 expression and the clinicopathological features and survival parameters were analyzed and prognostic factors were identified. PD-L1 expression was significantly associated with moderate/heavy smoking history and serum squamous cell carcinoma antigen (SCCA) levels. Multivariate analysis showed patients with high PD-L1 expression had significantly shorter disease free survival (DFS, HR 1.411, P = 0.025) and overall survival (OS, HR 1.659, P = 0.004) than those with low PD-L1 expression at a 50% cutoff value. No significant association was found between PD-L2 expression and patient postoperative survival. Further stratification analysis revealed that in patients with moderate/heavy smoking history, elevated serum SCCA level, and squamous cell carcinoma, PD-L1 expression was associated with significantly shorter DFS and OS. Therefore, PD-L1 expression was correlated with moderate/heavy smoking history and elevated serum SCCA level in NSCLC patients, and was an independently poor predictor of survival.
