Preclinical Safety and Pharmacokinetic Profile of Ferumoxtran-10, an Ultrasmall Superparamagnetic Iron Oxide Magnetic Resonance Contrast Agent

Abstract: The preclinical pharmacokinetic and safety profile of ferumoxtran-10 appears to be satisfactory in view of its proposed use as a single-dose diagnostic agent in human for MR imaging of lymph nodes.

“…Headache or back pain, vasodilation, hypotension, or allergic reaction in the form of urticaria can occur on rare occasions [62]. No systemic toxic effects have been observed in animal experiments at concentrations up to 100 mg Fe/kg [63]. The amount of a chronic iron overload (> 20 g) is not reached even when used multiple times.…”

“…133 In addition, another report shows the elimination of ∼20% of the injected dextran-coated SPIONs (with different size in comparison with ref 133) through urine and feces, in different animal models. 134 As predetermined, these differences in elimination of the NPs could be explained by variation of their protein corona compositions. Because of the physiological iron metabolism of cells, the surface degradation of SPIONs would be processed after their entrance to the intercellular medium, resulting in a temporary increase in iron amounts in the serum.…”

“…This indicates that USPIO incorporation leads to a more severe inflammatory response, but there were no signs of iron oxide-related toxicity and it is very unlikely that the scaffolds contain amounts beyond their safety level (2.6 mg Fe/kg). 31 Our assumption is this thicker capsule is due to more severe apparent inflammatory response by the introduction of iron particles 32 and the hypointensity because of iron particles residuals. Higher resolution and/or functional MRI could obtain more detailed information of the capsule.…”
