Precise transcriptional control of cellular quiescence by BRAVO/WOX5 complex in Arabidopsis roots

Abstract: Understanding stem cell regulatory circuits is the next challenge in plant biology, as these cells are essential for tissue growth and organ regeneration in response to stress. In the Arabidopsis primary root apex, stem cell-specific transcription factors BRAVO and WOX5 co-localize in the quiescent centre (QC) cells, where they commonly repress cell division so that these cells can act as a reservoir to replenish surrounding stem cells, yet their molecular connection remains unknown. Genetic and biochemical an… Show more

“…Additionally, FLIM is largely independent of protein concentration. Therefore, FRET-FLIM measurements are widely considered as more reliable than the detection of FRET by APB and spectral imaging, and have been successfully applied in planta ( Stahl et al , 2013 ; Bücherl et al , 2014 ; Somssich et al , 2015 ; Long et al , 2017 ; Weidtkamp-Peters and Stahl, 2017 ; Betegón-Putze et al , 2021 ; Kaur et al , 2021 ).…”

“…Ectopic expression of AtMYB56 resulted in the inhibition of root growth and the failure of root regeneration upon damage of stem cells [ 67 ]. Recently, the AtMYB56 protein has been reported to interact with the WOX5 protein to form a complex in QC cells ( Figure 2 B) [ 68 ]. In addition, an atypical 2R-MYB gene, At CDC5 ( cell division cycle 5 ), is well-known for its role in regulating the cell cycle.…”

“…This depends on the presence of long-lived stem cells, the capacity to reprogram somatic cells into pluripotent cells and the action of hormonal signals [ 126 ]. The response to hormonal signals is also directly linked to the activity of specific cell cycle regulatory factors, e.g., RBR1 in the auxin/cytokinin balance in the root [ 130 , 131 ], the ethylene response through ERF115 [ 132 , 133 ], the brassinosteroid signaling through BRAVO and CCS52A2 [ 134 , 135 , 136 ]. The interplay of hormonal signals with nutrient availability and metabolism is another field that should benefit from being analyzed from a cell proliferation perspective [ 137 , 138 ].…”
