Considering the necessity of Acrocomia aculeata propagation for large-scale production, the aim of this study was to establish a somatic embryogenesis protocol using the thin cell layer (TCL) technique. Aerial parts of in vitro plants were transversally cut at the base into eight TCLs and placed in a culture medium for callus induction. The induction medium was composed of Y3 salts and Morel´s vitamins and supplemented with 150, 300 or 600 μM picloram. After 12 weeks the calli were transferred to a medium supplemented with BAP or 2-iP (12.5 or 25 μM). After 18 weeks, the somatic embryo clusters were transferred to a conversion medium (plant growth regulator-free medium). Primary callus induction rate was higher in the first three TCLs and in media containing 150 or 300 μM picloram. The best maturation results were obtained in medium containing 12.5 μM 2-iP or 12.5 μM BAP. Few somatic embryos converted into plants. The histological analyses showed that callus induction started adjacent to vascular bundles after two days of culture, and somatic embryos arose in the periphery of nodular calli. This study showed that the TCL embryogenesis protocol is promising for in vitro multiplication of A. aculeata.

De novo organogenesis is a process through which wounded or detached plant tissues or organs regenerate adventitious roots and shoots. Plant hormones play key roles in de novo organogenesis, whereas the mechanism by which hormonal actions result in the first-step cell fate transition in the whole process is unknown. Using leaf explants of Arabidopsis thaliana, we show that the homeobox genes WUSCHEL RELATED HOMEOBOX11 (WOX11) and WOX12 are involved in de novo root organogenesis. WOX11 directly responds to a wounding-induced auxin maximum in and surrounding the procambium and acts redundantly with its homolog WOX12 to upregulate LATERAL ORGAN BOUNDARIES DOMAIN16 (LBD16) and LBD29, resulting in the first-step cell fate transition from a leaf procambium or its nearby parenchyma cell to a root founder cell. In addition, our results suggest that de novo root organogenesis and callus formation share a similar mechanism at initiation.

Peach palm (Bactris gasipaes Kunth.) cropping is an excellent alternative to native species exploitation; nevertheless, the problems with seed germination and conventional propagation justify the use of in vitro culturing. Aiming to asses TDZ pulsing effect on B. gasipaes morphogenesis, explants obtained from unarmed microplants were maintained in two treatments, half of them in MS free medium (without growth regulator) and the other half in MS with TDZ (0.36 μM). Both groups were transferred to growth regulator-free MS medium following 14 days of culture. After 84 days of culture, TDZ pulsing increased the growth and development of the shoots, restricted the growth and development of the roots, with no influence on adventitious bud induction or somatic embryogenesis. Furthermore, development of prickles, thickening of roots and chlorotic leaves were noted under TDZ pulsing. Leaf sheath histological analysis showed an epidermal origin and no vascularization of these prickles.

Abstract: Our results indicate that the use of growth regulators in microplants can lead to habituation and to different morphogenic pathways leading to potential niche establishment, depending on the positioning of the competent cells and their interaction with neighboring cells.
