PPARγ Agonists Attenuate Palmitate-Induced ER Stress through Up-Regulation of SCD-1 in Macrophages

Abstract: BackgroundClinical trials have shown that treatment of patients with type 2 diabetes with pioglitazone, a peroxisome proliferator-activated receptor (PPAR)γ agonist, reduces cardiovascular events. However, the effect of PPARγ agonists on endoplasmic reticulum (ER) stress that plays an important role in the progression of atherosclerosis has not been determined. We sought to determine the effect of PPARγ agonists on ER stress induced by palmitate, the most abundant saturated fatty acid in the serum. Methods and … Show more

“…This protective role of desaturation has been indeed demonstrated in some in vitro studies and further supported by the findings in SCD1 muscle transgenic mice . It is to be noted that SCD1 knock‐out mice are resistant to diet‐induced weight gain and obesity‐related metabolic disease , but it is due to a defective synthesis of TGs and cholesteryl esters in the lean animals and does not contradict the importance of acyl‐CoA desaturation in the cells under the pressure of high FFA levels.…”

“…In addition, we observed an increase in FAS mRNA and a tendency to increase SREBP1 and SCD1 in HFT RG KO mice. The activation of SCD1 in macrophages by rosiglitazone is an important pathway to reduce endoplasmic reticulum stress, thus decreasing cellular apoptosis, and depletory events (Ikeda et al, ). Furthermore, we showed an increase in PGC‐1α gene expression, a PPAR co‐activator which regulates genes involved in energetic metabolism and mitochondrial biogenesis (Puigserver and Spiegelman, ).…”

“…The relative mRNA levels were analyzed via the quantitative comparative Ct method and normalized with 18s gene expression as an internal control (Ikeda et al. ). The nucleotide sequences of primers are used:…”
