PP‐22 promotes autophagy and apoptosis in the nasopharyngeal carcinoma cell line CNE‐2 by inducing endoplasmic reticulum stress, downregulating STAT3 signaling, and modulating the MAPK pathway

Abstract: Paris polyphylla var. yunnanensis, named Chong Lou, is considered an antitumor substance. In this study, we investigated the effect of PP-22, a monomer purified from P. polyphylla var. yunnanensis, on the nasopharyngeal carcinoma cell line CNE-2 in vitro. The results showed that PP-22 could inhibit the proliferation of CNE-2 cells via the induction of apoptosis, with evidence of the characteristic morphological changes in the apoptosis in the nucleus and an increase in Annexin V-positive cells. In addition, we… Show more

“…The ER stress response exists in diverse biological systems and plays roles in the regulation of cell proliferation in many tumor cells . There is also evidence that the induction of ER stress proteins plays a role in apoptosis in various cancer cells . GRP78 functions as a molecular chaperone and is a major regulator of ER stress, as it maintains the integrity of the ER and controls the activation of unfolded protein response (UPR) signaling molecules .…”

“…Several studies have reported the induction of endoplasmic reticulum (ER) stress during apoptosis in various tumor cells. 17 Therefore, we investigated ER stress-associated proteins. Our results indicate that NCTD significantly increased the expression of GRP78, ATF-4,…”

“…A recent study reported that PDZ binding kinase, a serine–threonine kinase, accelerates cell apoptosis by activating the MAPK signaling pathway in nasopharyngeal carcinoma . PP‐22 promotes apoptotic and autophagic cell death by inducing endoplasmic reticulum stress and modulation of the p38 MAPK pathway in nasopharyngeal carcinoma CNE‐2 cells . Early reports have indicated that MAPK and PI3K pathways are targets for the development of therapeutic agents against nasopharyngeal carcinoma .…”

“…Hara are the two legal species according to the CP 2015 edition [10], which is the main raw material of Chinese medicine including "Yunnan Baiyao" and "Gongxuening". Studies have shown that PR has diverse pharmacological effects including antitumor [11][12][13], antimicrobial [14], anthelmintic activities [15], promoting hemostasis [16], antifungal [17], and immuno-stimulating properties [18]. The major active components in PR are spirostanol saponins including polyphyllin VII, VI, I, II, which are as the markers for quality control of PR [19].…”
