Potentiating photodynamic therapy of ICG-loaded nanoparticles by depleting GSH with PEITC

Abstract: In this work, we rationally combined novel ICG@HES-OA NPs with PEITC for potent PDT. The combination of ICG@HES-OA NPs and PEITC results in synergistic PDT efficacy.

“…It has also been shown that PEITC has an anticancer mechanism for a variety of cancers. PEITC binds to GSH to form GSH conjugates under the action of GSH S‐transferases, resulting in the depletion of GSH in cells, thereby eradicating cancer cells . Our results indicate that high doses of PEITC (10 µM) can significantly deplete intracellular GSH levels (Figure B).…”

“…Recently, Yang's group combined ICG-encapsulated nanoparticles with β-Phenylethyl isothiocyanate for potent PDT. [47] After encapsulation, the stability of ICG was enhanced and 1 O 2 was generated efficiently under 808 nm laser irradiation. The nanocomposite could consume intracellular glutathione and consequently boost the PDT efficacy.…”

“…Besides, ICG is a Food and Drug Administration (FDA) clinically approved NIR PS for PDT. Recently, Yang's group combined ICG‐encapsulated nanoparticles with β‐Phenylethyl isothiocyanate for potent PDT . After encapsulation, the stability of ICG was enhanced and 1 O 2 was generated efficiently under 808 nm laser irradiation.…”

“…Super carbonate apatite-ICG NPs improved ICG uptake in tumor cells and its antitumor effect in a colorectal xenograft model, serving as a useful vehicle for ICG-based PDT (Tamai et al, 2018 ). Also, hydroxyethyl starch-oleic acid ICG-NPs exhibited excellent stability and efficient ROS generation and increased cellular uptake and tumor accumulation compared to free ICG (Hu et al, 2019 ).…”
