Potential role of β-elemene on histone H1 in the H22 ascites hepatoma cell line

Abstract: Abstract. β-elemene is extracted from the Chinese medicinal herb Curcuma Wenyujin. It has a broad-spectrum antitumor effect on many types of cancer. However, the exact mechanism of action of β-elemene in hepatocellular carcinoma (HCC) remains unknown. Histone H1 is thought to act as a repressor of transcription by promoting the compaction of chromatin into higher order structures. We found that histone H1 plays an important role in the antitumor function of β-elemene. In this study, histone H1 expression in th… Show more

“…Curcuma zedoaria , the dried rhizome of Zingiberaceae plants, has previously been revealed to exhibit antitumor, antithrombotic and antibacterial effects, and has been used to treat many types of cancer in traditional Chinese medical practices ( 14 ). β-elemene, an extractable monomer from Curcuma zedoaria oil, has a wide antineoplastic spectrum, without toxic effects, and has been approved as an auxiliary treatment in China ( 8 ). Previous studies have revealed that the anticancer effects of β-elemene are predominantly due to the inhibition of tumor cell proliferation at a certain stage in the cell cycle, in addition to increased levels of apoptosis ( 7 9 ).…”

“…Similarly, most experimental studies of β -elemene targeting hepatobiliary diseases have been related to cancer. In mice models transplanted with human HCC cell lines (H22 [ 76 ] and MHCC97H cells [ 77 ]), β -elemene reduced the weight and volume of the tumor tissue. In particular, β -elemene enhanced the sensitivity of the anticancer drugs by increasing the level of copper transporter 1, aiding in the uptake of oxaliplatin into liver cells [ 77 ].…”

“…In particular, β -elemene enhanced the sensitivity of the anticancer drugs by increasing the level of copper transporter 1, aiding in the uptake of oxaliplatin into liver cells [ 77 ]. Based on the underlying mechanisms of its anticancer effects, β -elemene inhibited the proliferation of cancer cells by increasing histone H1 protein in H22 cells [ 76 ], and its treatment induced G2/M arrest and apoptosis of HepG2 cells by augmenting Fas and FasL mRNA and proteins on the cellular surface [ 75 ].…”
