Potential role ofFoxO1 and mTORC1 in the pathogenesis ofWestern diet‐induced acne

Abstract: Acne in adolescents of developed countries is an epidemic skin disease and has currently been linked to the Western diet (WD). It is the intention of this viewpoint to discuss the possible impact of WD-mediated nutrient signalling in the pathogenesis of acne. High glycaemic load and dairy protein consumption both increase insulin/insulin-like growth factor-1 (IGF-1) signalling (IIS) that is superimposed on elevated IGF-1 signalling of puberty. The cell's nutritional status is primarily sensed by the forkhead b… Show more

“…While human sebocytes may also express FoxO1, few relevant data exist until now [71,72]. In adipocytes, FoxO1 directly interacts with PPARγ to inhibit adipocyte differentiation and substantial evidence indicate a pivotal role FoxO1 in lipid homeostasis, regulation of oxidative stress, cell differentiation and apoptosis [70].…”

“…In adipocytes, FoxO1 directly interacts with PPARγ to inhibit adipocyte differentiation and substantial evidence indicate a pivotal role FoxO1 in lipid homeostasis, regulation of oxidative stress, cell differentiation and apoptosis [70]. It has been proposed that acne and increased sebaceous lipogenesis are associated with a relative nuclear deficiency of FoxO1 caused by increased growth hormone (GH)/insulin/IGF-1/or FGFR2 signaling [72].…”
