Potential Role of OCT4 in Leukemogenesis

Abstract: Embryonic stem cells typically show properties of long-term self-renewal and lack of differentiation. When appropriately stimulated, they are able to differentiate into all cell lineages, and lose their self-renewal characteristics. These properties are controlled by a series of genes encoding several transcription factors, including OCT4, the product of POU5F1 gene. OCT4 is expressed in germ cell tumors but also aberrantly in cancers developing in differentiated tissues. In a previous study, we observed a hig… Show more

“…51 In agreement, we showed that cell cycle arrest and mitophagy induced by CBD in CML cells are associated with changes in the differentiation markers OCT-4 and PU.1 involved in leukemogenesis and myeloid differentiation. 52,53 We found a reduction of the stem-associated marker OCT-4 level and the enhancement of PU.1, suggesting that CBD drives CML cells to a more differentiated state. This was also confirmed by colony formation assay that demonstrates the ability of CBD treatment to reduce the clonogenic activity of CML cells.…”

“…3c). Among these factors, LYL1 has oncogenic potential in AML 54 while POU5F1 (also known as OCT4) was implicated in differentiation arrest and proliferation mechanisms through oncogenic mechanisms 55 . Our results suggest that additional transcription regulators, namely NFYA and NFYB, contribute to the stemness properties typical of LSCs in AML.…”

“…Inhibiting the expression of the OCT4 gene prolongs the cell division time and also reduces the proliferation of cancer cells (29). The inhibition of the expression of the OCT4 gene causes the cell cycle to stop, thereby reducing the proliferative capacity and creating a colony in the acute myeloid cell line (30). Decreased OC4 expression in colorectal metastatic cancer patients who responded well to treatment was observed, of which OCT4 could be used to predict response to treatment in this malignancy (31).…”
