Potential of bone scaffolds containing vancomycin and bone morphogenetic protein-2 in a rat model of osteomyelitis

Abstract: Background: Infected bone is often intractable. An ideal approach is to simultaneously eradicate infection and repair the bone defect. The development of osteoinductive bone graft composites to control antibiotic drug release would be useful for the treatment of intractable bone infections. Objectives: To develop a rat model of osteomyelitis for assessing osteoinductive bone graft scaffolds containing antibiotics and a bone morphogenetic protein. Methods: Si-imprinted calcium phosphate is a new hydroxyapatite d… Show more

“…The most frequently used antibiotics in local drug delivery systems are vancomycin (VAN) [18,20,21,27,33,42,48,53,[60][61][62][63]65] and gentamicin (GT) [18,23,25,27,35,46,[57][58][59]65]. These are also the most ubiquitously applied antibacterial agents in systemic therapy of bone tissue infections.…”

“…Moreover, vancomycin and the BMP-2 double-drug-loaded silicon-imprinted calcium phosphate scaffold and BMP-2 single-drug-loaded silicon-imprinted calcium phosphate scaffold both significantly promote new bone formation of rat tibial defects. The amount of new bone formation between the two is comparable, suggesting that vancomycin with appropriate dose would not damage the osteogenic induction ability of BMP-2 [ 189 ]. Similarly, vancomycin can also play a good antibacterial role in this dual drug loading system.…”
