Potential mechanism of action of J5 vaccine in protection against severe bovine coliform mastitis

Abstract: -Coliform mastitis is one of the most difficult diseases to treat in the modern dairy industry. Curative therapy with antibiotics remains only moderately effective and depends on the stage at which the disease is treated. The most successful strategies for combating coliform mastitis appear to be prevention by hygienic management or prophylactic immunization. The severity of clinical symptoms of coliform mastitis has been shown to be reduced by immunization with the Escherichia coli J5 vaccine. However, althou… Show more

“…This is related to the hypothesis of Dosogne et al (2002) that the J5 vaccination might induce a T helper 1 (Th 1 ) response. Indeed, the Th 1 response is characterized by the production of cytokines that enhance PMN diapedesis, as reflected in the high υ values.…”

“…E. coli serotypes in mastitic milk are similar to the faecal isolates. There are a very large number of E. coli serotypes as classified by surface antigens but the inner layer of the cell wall is common to all serotypes of E. coli and Enterobacteriaceae in general [65].…”

“…During multiplication, destruction and lysis, they release an outer-membrane component, the endotoxin or LPS [68], a thermostable molecule that is a unique structural component of the cell wall of the Gram-negative pathogens. Two phenotypic forms of coliforms with slightly different LPS can be distinguished: smooth with complete LPS sidechains and rough with incomplete LPS side-chains [65,154,169]. Many studies have shown that the pathogenicity of E. coli in the mammary gland is associated with LPS.…”

“…Several attempts have been made to prevent cows from contracting E. coli mastitis through vaccination [40]. E. coli strain J5 is a rough (Rc) mutant strain that consists of a conserved lipid A portion of the cell wall and some common core polysaccharides.…”
