Potato cyst nematodes (Globodera rostochiensis and G. pallida) from Swedish potato cultivation - an AFLP study of their genetic diversity and relationships to other European populations

Abstract: Sixteen Swedish PCN populations (nine Globodera rostochiensis and seven G. pallida populations) were compared with 20 other European populations (nine G. rostochiensis and 11 G. pallida populations) from Germany, the Netherlands, Norway and the United Kingdom in an AFLP study. Seven AFLP primer combinations were used to amplify genomic DNAs extracted from cysts. A dendrogram, based on UPGMA and evaluated by bootstrap analysis, contained two main clusters, one for each species. The Swedish Ro1 populations were … Show more

“…The wide divergence found between two species, G. pallida and G. rostochiensis , is in agreement with previous studies on the basis of two‐dimensional gel electrophoresis (2‐DGE) (Bossis and Mugniéry 1993), RLFP (Gonzalez et al. 1997), AFLP (Manduric and Andersson 2003) and SSR primers (Blok and Philips 1995). Populations belonging to different Globodera spp were studied on several occasions.…”

“…Before the use of molecular markers, most hypotheses were poorly supported by any tangible data, or based only on the geographical distribution of resistance genes among wild potatoes (Franco 1977). Although a range of studies found intraspecific polymorphism in European populations of G. pallida for various molecular markers, they either did not include any Peruvian populations (Phillips et al 1992;Folkertsma et al 1994;Burrows et al 1996;Folkertsma et al 1996aFolkertsma et al , b, 2001Thiéry & Mugniéry 1996;Manduric & Andersson 2003) or failed in precise identification of the origin of European populations due to the low representation of Peruvian samples (Blok & Phillips 1995;Blok et al 1997Blok et al , 1998Subbotin et al 2000;Grenier et al 2001).…”

“…The four populations, previously characterised in an AFLP study (Manduric & Andersson, 2003) were first reared on potatoes cv. Bintje in a glasshouse at a temperature of about 20 • C. The progeny was then inoculated into an artificial soil mixture (70% silver sand, 20% clay powder, 10% humus) in 1 l pots with potato cv.…”
