Posterior rat eye during acute intraocular pressure elevation studied using polarization sensitive optical coherence tomography

Abstract: Polarization sensitive optical coherence tomography (PS-OCT) operating at 840 nm with axial resolution of 3.8 µm in tissue was used for investigating the posterior rat eye during an acute intraocular pressure (IOP) increase experiment. IOP was elevated in the eyes of anesthetized Sprague Dawley rats by cannulation of the anterior chamber. Three dimensional PS-OCT data sets were acquired at IOP levels between 14 mmHg and 105 mmHg. Maps of scleral birefringence, retinal nerve fiber layer (RNFL) retardation and r… Show more

“…Furthermore, the effects may differ with age. 47,48 Different metrics, such as retinal layer thickness, 49 retinal capillary density, 14 birefringence, 50 or gene expression, 51 can be affected to different degrees by IOP elevation. We believe that vis-OCT, which has the ability to provide structural information, as well as data on perfusion of major retinal vessels and capillary beds and total retinal blood flow, oximetry, oxygen extraction, and oxygen metabolism, offers several advantages that may help in understanding these phenomena.…”

“…In this work we utilized a multifunctional OCT imaging system that was described in detail by Fialová et al 43 and has been recently used to image mouse 41 and rat models 42,44 of ophthalmic diseases. In short, the system is based on a SD, PS-OCT unit enabling high-speed retinal imaging at 83-kHz Ascan rate with an axial resolution of 3.8 lm in retinal tissue.…”

“…ONH and lamina cribrosa biomechanics are also often studied using optical coherence tomography, which has the important advantage that it can be used in vivo. 1,41,42 However, despite exciting advances, such as the integration of polarized light information, 43,44 the light-penetration, resolution, and contrast of these systems are not yet sufficient to quantitatively analyze lamina cribrosa beams with the level of detail demonstrated with SPLM in this work. A study using serial sectioning of eyes fixed at various intraocular pressures followed by PLM imaging has addressed some of these issues, and indeed provided valuable findings.…”
