Asian Pac J Cancer Prev, 15 (16), 6535-6541 IntroductionBreast cancer related lymphedema (BCRL)is a chronic and common complication caused by abnormal accumulation of protein-rich fluid in the interstitial space secondary to inadequate lymphatic drainage, which manifests unilateral or bilateral upper extremity swelling in the clinical. (Cheville et al., 2003). Lymphedema and its related symptoms (pain, heaviness, tightness, and decreased range of motion) seriously affect recreational and social relationships (Hayes et al., 2012). Patients with BCRL have a lower quality of life, a higher level of anxiety or depression (Pyszel et al., 2006;Heiney et al., 2007).Due to differences in study designs, measurement methods and criterias , length of follow-up, lymphedema definitions and timing of lymphedema measurement since diagnosis and treatment, previous studies have shown about 20% of breast cancer survivors will develop lymphedema. (sakorafas et al., 2006) In recent years, many studies have established risk factors of BCRL, but conclusions are not consistent, which even contradict each other from some studies. The study systematically reviews the risk factors for BCRL with meta-analysis method to increase the credibility of the conclusions for the purpose of providing scientific evidence for early prevention of lymphedema in clinical work. Materials and Methods Literature SearchWe searched comprehensively clinical studies which were published in PubMed, Ovid, EMbase, the Cochrane Library from January 1, 1996 to December 30, 2012. The MeSH headings and keywords in this search used included "breast cancer", "upper extremity", "lymphedema" and "risk factors" Relevant articles were identified and their references were checked for additional studies.The search strategy used: ( ("Breast Neoplasms" (Mesh) AND "Upper Extremity" (Mesh)) AND ("Lymphedema" (Mesh) OR "Edema" (Mesh)) AND ("1996/01/01" (PDAT) : "2012/12/31" (PDAT)) Inclusion and Exclusion CriteriaInclusion criteria: studies of female patients with unilateral breast cancer; the primary studies of risk factors for BCRL published abroad.Exclusion criteria: studies of patients with bilateral breast cancer, primary lymphoedema, or metastatic disease (n=258); review, meta-analyses, editorial, comment , and case reports (n=121); studies that did not either inform OR and 95%CI or provide adequate information to calculate the OR and its variance (n=39); non-English-language studies (n=8).

Background: Nigella Sativa (NS) is an herb from the Ranunculaceae family that exhibits numerous medicinal properties and has been used as important constituent of many complementary and alternative medicines (CAMs). The ability of NS to kill cancer cells such as PC3, HeLa and hepatoma cells is well established. However, our understanding of the mode of death caused by NS remains nebulous. The objective of this study was to gain further insight into the mode and mechanism of death caused by NS in breast cancer MCF-7 cells. Materials and Methods: Human breast cancer cells (MCF-7) were treated with a methanolic extract of NS, and a dose-and time-dependent study was performed. The IC 50 was calculated using a Cell Titer Blue ® viability assay assay, and evidence for DNA fragmentation was obtained by fluorescence microscopy TUNEL assay. Gene expression was also profiled for a number of apoptosis-related genes (Caspase-3, -8, -9 and p53 genes) through qPCR. Results: The IC 50 of MCF-7 cells was 62.8µL/mL. When MCF-7 cells were exposed to 50 µL/mL and 100 µL/mL NS for 24h, 48h and 72h, microscopic examination (TUNEL assay) revealed a dose-and time-dependent increase in apoptosis. Similarly, the expression of the Caspase-3, -8, -9 and p53 genes increased significantly according to the dose and time. Conclusions: NS induced apoptosis in MCF-7 cells through both the p53 and caspase pathways. NS could potentially represent an alternative source of medicine for breast cancer therapy.

INTRODUCTIONHuman DDX3 gene encodes a polypeptide of 662 amino acids and this protein plays an important role in several aspects of RNA metabolism. 1,2 This protein has two clear distinguishable domains comprised of N-terminal DEAD box domain 1 (211-403 residues) and C-terminal helicase domain 2 (411-575 residues). Both domains displayed RecA-like folds comprising a central β-sheet flanked by α-helices connected by a noncanonical linker of 11 amino acids.3 Expression of DDX3 was detected in several tissues such as testis, colon, lung, liver, skeletal muscle, and kidney 2,4 indicating the universal role of DDX3 in cellular homeostasis. Moreover, elevated expression of DDX3 was found in a highly aggressive metastatic breast cancer cell line, MDA-MB-231, as compared with non-metastatic MCF-7 cells, which show its potential role in aggressive breast cancers and associated metastasis. 5,6 We have previously reported that overexpression of DDX3 in immortalized non-turmorigenic MCF10A cells promoted neoplastic transformation as indicated by down regulation of a cell adhesion molecule, E-cadherin.5 Down-regulation of E-cadherin is a common feature of a variety of metastatic epithelial tumors, including those of the lung, breast and prostate cancer.5,7-9 Hypoxic regions of solid tumors were considered to be the primary sites for the generation of the metastatic phenotype and have been demonstrated to be chemo and radio-resistant. [10][11][12][13][14] We have also reported that hypoxia inducible factor (HIF-1) induce the expression of DDX3 in two different breast cell lines by binding directly or indirectly to the hypoxia-response element (HRE) in the DDX3 proximal promoter. 15 On the other hand, a significant down regulation of DDX3 expression is found in hepatocellular carcinoma (HCCs) from the hepatitis B virus (HBV) positive patients, but not from HCV positive in comparison to the corresponding non tumor tissues. 16 In the hepatocellular carcinoma model, DDX3 was found to act as a tumor suppressor by activating the expression of cyclin dependent kinase inhibitor p21 cip1 . 17 Besides cancer, induced expression of DDX3 was also found in HIV-1 infected cells. 18,19 Overall, it suggests that DDX3 is a multifunctional protein and it plays a specific role in regulatory mechanisms and signaling pathways. Apart from embryonic development, this gene is also involved in multiple diseases like HIV, Neuro-degenerative diseases, hepatocellular carcinoma and brain and breast cancer.
