Positive inotropic, positive chronotropic and coronary vasodilatory effects of rat amylin: mechanisms of amylin-induced positive inotropy

Abstract: Even though there are a few studies dealing with the cardiac effects of amylin, the mechanisms of amylin-induced positive inotropy are not known well. Therefore, we investigated the possible signaling pathways underlying the amylin-induced positive inotropy and compared the cardiac effects of rat amylin (rAmylin) and human amylin (hAmylin).Isolated rat hearts were perfused under constant flow condition and rAmylin or hAmylin was infused to the hearts. Coronary perfusion pressure, heart rate, left ventricular d… Show more

“…Direct cardiac inotropic effects, potentially CGRP receptor mediated, have been reported for amylin in isolated rodent cardiomyocytes (Bell and McDermott, 1995), isolated rat hearts (Kaygisiz et al, 2010), and isolated cardiac tissue from pigs (Saetrum Opgaard et al, 1999). However, because of the high concentrations required to observe efficacy, it is again unlikely that these actions are clinically relevant at therapeutic doses.…”

“…Studies on rat cardiomyocytes and isolated heart, AMY showed a direct inotropic effect that mediated by CGRP receptors. However, because of the high doses AMY application side-effects on the heart, it was discussed that AMY could not be applied to the clinic 44,45 .…”
