The aim of study was to scrutinize the physicochemical and protein profile of milk obtained from local Pakistani breeds of milch animals such as Nilli-Ravi buffalo, Sahiwal cow, Kajli sheep, Beetal goat and Brela camel. Physicochemical analysis unveiled maximum number of total solids and protein found in sheep and minimum in camel. Buffalo milk contains the highest level of fat (7.45%) while camel milk contains minimum (1.94%). Ash was found maximum in buffalo (0.81%) and sheep (0.80%) while minimum in cow’s milk (0.71%). Casein and whey proteins were separated by subjecting milk to isoelectric pH and then analyzed through sodium dodecyl sulphate-polyacrylamide gel electrophoresis (SDS-PAGE). The results showed heterogeneity among these species. Different fractions including α S1 , α S2 , κ-casein, β-casein and β-lactoglobulen (β-Lg) were identified and quantitatively compared in all milk samples. Additionally, this electrophoretic method after examining the number and strength of different protein bands (α S1 , α S2 , β-CN, α-LAC, BSA, and β-Lg, etc.), was helpful to understand the properties of milk for different processing purposes and could be successfully applied in dairy industry. Results revealed that camel milk was best suitable for producing allergen free milk protein products. Furthermore, based on the variability of milk proteins, it is suggested to clarify the phylogenetic relationships between different cattle breeds and to gather the necessary data to preserve the genetic fund and biodiversity of the local breeds. Thus, the study of milk protein from different breed and species has a wide range of scope in producing diverse protein based dairy products like cheese.

El objetivo de este estudio fue evaluar la diversidad genética de siete poblaciones de bovinos para carne en México, mediante análisis de sus pedigríes. Los análisis se realizaron con el programa Endog, utilizando información de los animales inscritos en el libro genealógico nacional de cada raza. Los tamaños de los pedigríes estudiados fueron: Angus (AN)= 73,271; Brangus Negro (BN)= 68,474; Brangus Rojo (BR)= 12,925; Hereford (HE)= 13,248; Limousin (LI)= 53,221; Salers (SA)= 14,065; y Suizo Europeo (SE)= 184,788. En general, las poblaciones mostraron mejoras importantes en la integridad de sus pedigríes en los 10 años más recientes, con parámetros comparables a los publicados en otras poblaciones de bovinos. El intervalo generacional varió de 5.1 a 7.2 años entre razas. Los coeficientes de consanguinidad y de relación genética aditiva promedio fueron relativamente bajos en las diferentes poblaciones (0.9-4.2 y 0.3-6.5 %, respectivamente); aunque en algunas razas (BN, BR y HE) se detectaron incrementos de estos indicadores en los 10 años más recientes. El tamaño efectivo de población fluctuó entre 24 y 192, con valores menores que 50 en BR y SA. Los parámetros relacionados con la probabilidad de origen de los genes indican que las contribuciones desbalanceadas de los animales fundadores, los cuellos de botella y la deriva genética, han tenido un efecto importante en las poblaciones actuales, con la consecuente pérdida de diversidad genética. Se recomienda continuar o adoptar estrategias de apareamiento que minimicen la consanguinidad, y el uso intensivo de pocos animales para mantener la variabilidad genética de futuras generaciones, así como ampliar la diversidad genética mediante el uso estratégico de genes provenientes de otros países.

Breeding and genetics Full-length research article Population structure and inbreeding of Holstein cattle in southern Brazil ABSTRACT-The present investigation aimed to evaluate the population structure and inbreeding of Holstein herds in southern Brazil. To carry out the analysis, the Associação Paranaense de Criadores de Bovinos da Raça Holandesa (APCBRH) in Brazil provided the data, which consisted of a pedigree file of 206,796 animals born between 1970 and 2014. Results regarding the following parameters were determined: pedigree integrity, effective number of founders, effective number of ancestors, generation interval, inbreeding coefficient, realized effective population size, and average relatedness coefficient. POPREP and ENDOG v.4.5 software packages were employed to estimate these parameters. Based on the data set, the mean generation interval was found to be 6.3 years, and the average inbreeding coefficient, related to inbred animals, was 4.99%. Furthermore, the realized effective population size varied throughout the time period, ranging from 22 to 114, whereas the rate of inbreeding in this same period showed a decreasing trend towards the later years in the period until 2014. Upon evaluation, average relatedness coefficient was estimated to be 0.71%. Moreover, the effective number of founders and ancestors were estimated as 418 and 400 animals, respectively. According to the level of inbreeding observed, it could be noticed that genetic diversity remains elevated, which will be important to the genetic progress in the Holstein breeding program in Southern Brazil.
