Pontimonas salivibrio gen. nov., sp. nov., a new member of the family Microbacteriaceae isolated from a seawater reservoir of a solar saltern

Abstract: Pontimonas salivibrio gen. nov., sp. nov., a new member of the family Microbacteriaceae isolated from a seawater reservoir of a solar saltern A Gram-staining-positive, non-motile, strictly aerobic, non-spore-forming, vibrio-shaped bacterial strain, CL-TW6 T , was isolated from a reservoir seawater sample from a solar saltern in Korea. Analysis of the 16S rRNA gene sequence of strain CL-TW6 T revealed a clear affiliation with the family Microbacteriaceae. Strain CL-TW6 T showed the closest phylogenetic relation… Show more

“…The Actinobacteria clones were most related (90.5%–99.7%) to environmental sequences from a Tibetan lake (Zhang et al, ). The closest cultured relative was Pontimonas salivibrio isolated from the seawater reservoir of a solar saltern in Korea (Jang, Cho, & Cho, ) and the freshwater genus “ Candidatus Planktoluna” (Hahn, ). The Bacteroidetes clones affiliated with phyla Saprospira and Flavobacteriia, two of which were closely related to clone sequences from a Tibetan lake (Zhang et al, ).…”

“…1). This freshwater cluster including strain MWH-Ta8 T appeared in all trees calculated by the three methods together with Pontimonas salivibrio CL-TW6 T (Jang et al , 2013). The close relationship of these taxa was supported with high bootstrap values by at least two methods, but the phylogenetic relationship of the Pontimonas /Luna-1 cluster to other genera of the family Microbacteriaceae could not be resolved.…”

“…The pH range for growth was also determined by assessing changes in the OD 600 in MB adjusted to pH 5.0-10.0 (in increments of 0.5 pH units), incubated at 35 u C for 7 days. The following buffers were used for pH experiments: citrate/phosphate (pH 5.0 and 5.5), MOPS (Sigma) (pH 6.0-8.0), boric acid/borax (pH 8.5 and 9.0), carbonate/bicarbonate (pH 9.5 and 10.0), borax/NaOH (pH 9.5 and 10.0) and Tris/HCl (pH 8.5-10.0), each at a final concentration of 50 mM (Scheidle et al, 2011;Jang et al, 2013). The pH of a subsample of autoclave-sterilized medium was measured before inoculation of cells to check for changes in pH.…”
