Polyribosomes from Peas

Abstract: Profiles of polyribosomes were obtained from etiolated stem segments of Pisum sativum L. var. Alaska isolated in various buffers. Tissue homogenized in a medium containing 0.2 M tris-HCI, pH 8.5, 0.2 M sucrose, 30 mM MgCL, and 60 mM KCI yielded polyribosomes exhibiting far less degradation than tissue homogenized in conventional media containing tris-HCl at lower ionic strength and pH. A further decrease in degradation was found when polyribosomes were sedimented through a sucrose pad buffered at pH 8.5 prior … Show more

“…An analysis of the profiles in Figure 3 is presented in Table I. The ratio of the large polysomes to the total polysomes has previously been used as an indication of the relative degradation of polysomes (6,11 Varying Mg2+ Concentrations Maize kernels were ground in buffer A containing 1 mM Mg2+. The homogenate was strained through four layers of cheesecloth and centrifuged at 5OOg for 5 min.…”

“…The methods described by Davies et al (9) for pea stem internodes, Lin et al (22) for soybean roots, and Bewley (3) for moss also resulted in poor yields of the polysome fraction with a high yield of free ribosomal material. The unmodified technique of Jachymczyk and Cherry (18) produced yields only slightly lower than the present method.…”

“…Isolation of RNA. The extraction procedure was modified from Davies et al (13), with heparin addition based on Palmiter (37). In a typical extraction, frozen kernels were cooled in liquid N2, 2 Reference to a company and/or product named by the department is only for purposes of information and does not imply approval or recommendation of the product to the exclusion of others which may also be suitable.…”
