“…Furthermore, inositol polyP has been suggested to have numerous roles in signaling and developmental process during germination, including gene expression (Shears 2004;Bowen et al 2007). The globoid is suggested to be involved in embryo development during seed germination, like the acidocalcisome-like organelle found recently in the egg yolks of insects (Gomes et al 2008). In insect eggs, the acidic environment of the matrix induces polyP synthesis, which promotes Ca storage.…”

“…In insect eggs, the acidic environment of the matrix induces polyP synthesis, which promotes Ca storage. This process acts as a dynamic system for P i and Ca homeostasis (Gomes et al 2008). It is possible that a similar mechanism operates in seeds.…”

“…23 DAPI-PolyP interaction was shown to be remarkably specific and to generate a proper quantum yield for microscopic observation. 21,24-26 Nevertheless, reports concerning the feasibility of using DAPI as a PolyP probe have remained fragmented among several publications that suggested a number of possible routines for DAPI-PolyP labeling and visualization. 23,27,28 …”

“…The presence of poly P stored in acidocalcisomes has been extensively reported in different models [15], [23], and changes in its levels according to varied extracellular stimuli were observed [35]. Interestingly, the levels of poly P in Rhodnius acidocalcisomes tend to decrease slightly before the beginning of yolk mobilization, and a decrease in the levels of this polymer was already observed in eggs of the cattle tick Boophilus microplus [61] and the cockroach P. americana [37]. These data suggest that poly P might be widely used as Pi source for growing embryos, mainly in invertebrates, that do not store massively phosphorylated yolk proteins as phosvitins, known in vertebrate models [62].…”

“…Total Poly P was extracted as described before by Gomes et al (2008) [37]. Acidocalcisome fractions were resuspended in water and sonicated (probe sonication, 30% amplitude, three cycles of 20 s).…”
