Polyphenol Extracts from Hibiscus sabdariffa Linnaeus Attenuate Nephropathy in Experimental Type 1 Diabetes

Abstract: Diabetic nephropathy progressed to end-stage renal disease (ESRD) is found in type 1 or type 2 diabetes. Oxidative stress is one of the precipitation factors in diabetic nephropathy. Previously, Hibiscus sabdariffa Linnaeus and its polyphenol extracts were found to possess antioxidative effects. This study is aimed to investigate the effect of Hibiscus sabdariffa L. polyphenol extract (HPE) in streptozotocin (STZ) induced diabetic nephropathy. The results show that HPE reduced kidney mass induced by STZ signif… Show more

“…Nevertheless, ethanol extracts of HSE supplementation decrease the protein carbonyl level and this is in agreement with the previous study, that Roselle effectively decrease protein oxidation in renal and hepatic tissues of diabetic rats (Farombi & Ige, 2007). In addition, polyphenols in H. sabdariffa was shown to reduce oxidative damage of kidney and improve diabetic nephropathy condition (Lee et al, 2009). Table 1 shows a significantly low activity of SOD and CAT as well as GSH (p<0.05) in the thoracic aorta of DM group compared to NDM group.…”

“…9 Most importantly, HS was recently demonstrated to be effective in attenuating the progression of kidney injury in streptozocin-induced diabetic nephropathy in rats through its antilipidemic and antioxidant properties. 15 As a result of this, we investigated the effects of HS using a 5/6 Nx rat model for the study of CKD as a proof of concept for early intervention therapy. HS was administered 1 week after 5/6 Nx.…”

“…Moreover, HS improves the conditions of streptozotocin-induced diabetic nephropathy in rat via several mechanisms such as antihypertensive action, lipid-lowering effect, and antioxidant properties. 15 Hence, the multiple effects of HS extract should be effective against multiorgan dysfunctions in CKD patients. However, there are no data on the effects of HS in CKD.…”

“…Administration of polyphenols to diabetic animals resulted in a lower level of markers of oxidative stress as judged by lower levels of products of lipid peroxidation in pancreatic homogenates (Coskun et al 2005), kidneys (Lee, Wang, et al 2009) and blood plasma (Ciocoiu et al 2009;Hininger-Favier et al 2009) of STZ-rats. This may be a result of direct antioxidant activity of polyphenols, as well as a result of their ability to upregulate expression of antioxidant-defense enzymes: glutathione peroxidase (GSHPx) superoxide dismutase and catalase activities (Ciocoiu et al 2009;Lee, Wang, et al 2009;Dixit and Kar 2010).…”

It is well known that oxidative stress plays a substantial role in the destruction of beta-cells by infiltrated self macrophages and lymphocytes.…”
