Polydatin prevents fructose-induced liver inflammation and lipid deposition through increasing miR-200a to regulate Keap1/Nrf2 pathway

Abstract: Oxidative stress is a critical factor in nonalcoholic fatty liver disease pathogenesis. MicroRNA-200a (miR-200a) is reported to target Kelch-like ECH-associated protein 1 (Keap1), which regulates nuclear factor erythroid 2-related factor 2 (Nrf2) anti-oxidant pathway. Polydatin (3,4′,5-trihydroxy-stilbene-3-β-D-glucoside), a polyphenol found in the rhizome of Polygonum cuspidatum, have anti-oxidative, anti-inflammatory and anti-hyperlipidemic effects. However, whether miR-200a controls Keap1/Nrf2 pathway in fr… Show more

“…Previous studies provided a robust evidence to support a cross-talk between Nrf2 and miR-200a [ 20 , 21 ]. Hence, we speculated that miR-200a could act as an upstream of Nrf2, and was closely involved in the effect of PIC in macrophages.…”

“…Total cellular protein was extracted using NP40 lysis buffer, and the protein content was measured using a BCA protein assay kit. The expression levels of the proteins were measured as described previously (Zhao et al, ). Total protein was fractionated on 7% SDS‐PAGE and transferred to a polyvinylidene fluoride membrane.…”

“…And we realized that the increase in miR‐200a was greater than others after KCNQ1OT1 knockdown in oxygen and glucose deprivation and re‐oxygenation (OGD/R) cells. MiR‐200a was determined to participate in inflammatory and senescence processes in cells (Zhao et al, ). In addition, miR‐200a inhibits proliferation in carcinoma cells (Li et al, ).…”
