Polycarboxylates enhance beetle antifreeze protein activity

Abstract: SummaryAntifreeze proteins (AFPs) lower the noncolligative freezing point of water in the presence of ice below the ice melting point. The temperature difference between the melting point and the noncolligative freezing point is termed thermal hysteresis (TH). The magnitude of the TH depends on the specific activity and the concentration of AFP, and the concentration of enhancers in the solution. Known enhancers are certain low molecular mass molecules and proteins. Here, we investigated a series of polycarbox… Show more

“…Trisodium citrate was reported as one of the strong enhancers of antifreeze activity in terms of both TH and ice crystal growth modification. 30 Citrate also causes the highest increase in the TH of insect AFPs. 28 The addition of citrate noticeably increased the activity of the DCR analogues (DCR26, DCR39, DCR26 cyclic and DCR39 cyclic), not only promoting extreme crystal shaping resembling the very small ice crystals produced by the native DcAFP protein, but also creating a significant increase in TH (Fig.…”

“…The Hoffmeister effect may be responsible for enhancement of TH by monovalent salts (Wang et al, 2009a). Increasing numbers of hydroxyl groups in a series of polycarboxylates (Amornwittawat et al, 2008) and polyhydroxy compound enhancers (Amornwittawat et al, 2009) positively affected the TH of DAFP-1. Arginine is a key residue in the enhancement (Wang et al, 2009b).…”
