Poly(hydroxy acid) Nanoparticles for the Encapsulation and Controlled Release of Doxorubicin

Abstract: Diblock poly(hydroxy acid) copolymers are created by the sequential ring‐opening polymerization of l‐phenylalanine O‐carboxyanhydride and l‐lysine(carboxybenzyl) O‐carboxyanhydride, and l‐phenylalanine O‐carboxyanhydride and γ ‐benzyl l‐glutamic acid O‐carboxyanhydride. Upon protecting group removal, two amphiphilic block copolymers are formed that can self‐organize in aqueous solution to form spherical nanoparticles. Such nanoparticles are capable of encapsulating doxorubicin before allowing its programmed pa… Show more

“…Dox loading into polymer nanoparticles was achieved by 'dropping in' a solution of polymer dissolved in DMF to vigorously stirred phosphate buffered saline (PBS) solution that contained Dox. 30 (Table S4). However, in this instance a second peak formed over time that, after 21 days, was representative of particles that possess a hydrodynamic diameter of 31 nm and represented 6.1% of the total sample measured.…”

“…Nanoparticles werep roduced from the three polymer types by coacervation. [15] DLS analysisr evealed increased nanoparticle size with an increased proportion of hydrophobic PBLG within the block co-polymer (Table 1). PDI values of PBLG 2 -b-PEG 113 and PBLG 26 -b-PEG 113 were less than, or close to 0.3, indicating the particle stability.P BLG 35 -b-PEG 113 nanoparticles were considered unstableafter 21 days of storagedue to the recorded PDI value (0.437).…”
