The U.S. federal government often devolves administrative processes and decision making to state and local governments. Prior studies have found that the success of decentralization and implementation depends on several mediating factors at the subnational level, such as a state's political ideology or administrative capacity. This study focuses on one mechanism—the ability of states to leverage their information advantage about the local context vis-à-vis the federal government. We are interested in whether the information advantage of state-level firearm background checks decreased the rates of intimate partner violence (IPV) involving a firearm compared to states that relied on federally administered background checks. We take advantage of data from the period of state-mandated stay-at-home (SAH) orders during the COVID-19 pandemic, a temporal context with increased IPV rates. Using a Poisson fixed effects regression, we find that rates of IPV involving a firearm did not increase when the SAH orders were in effect. However, using decision-relevant information in state-administered background checks decreased the rates of IPV resulting in injury and murder-suicide, compared to states that relied on federal background checks.

Foi estudada a segurança e eficácia de formulações intraoculares contendo Luteína e Zeaxantina, isolada ou combinada com Azul Brilhante ou Azul de Trypan em cirurgia de catarata e vitreorretiniana. Soluções-corantes contendo óleo de rícino polietoxilado (Cremophor EL™) como veículo, não apresentam afinidade corante para a Cápsula Anterior, Vítreo, Hialóide Posterior, Membrana Limitante Interna e Membrana Epirretiniana. Adicionalmente, as soluções contendo Cremophor EL™ mostraram-se citotóxicas. Pelo contrário, soluções-corantes contendo álcool polivinílico como veículo mostraram-se eficazes e seguras. Os testes de eficácia em olhos cadavéricos mostraram apenas cinco formulações (sem Cremophor EL™) que coraram o vítreo menos de +++/4. Quanto à Cápsula Anterior e Membrana Limitante Iinterna, apenas a formulação de Luteína e Zeaxantina a 0,5% associada a Azul de Trypan a 0,04%; e a Luteína e Zeaxantina a 0,3% associada a Azul Brilhante a 0,025% atingiram a máxima coloração com os níveis mais baixos de Azul de Trypan e Azul Brilhante. O estudo de toxicidade intravítrea mostrou que nenhuma formulação testada exibe toxicidade “in-vivo” no vítreo e retina. As soluções-corantes Luteína e Zeaxantina a 0,3%-1% associada a Azul Brilhante a 0,025% e Luteína e Zeaxantina a 0,5%-1% associada a Azul de Trypan a 0,04%, ambas em álcool polivinílico, foram as melhores fórmulas em termos de estabilidade, pH e osmolaridade. Estas formulações atingiram os melhores resultados na coloração da Cápsula Anterior, Vítreo e Membrana Limitante Interna de olhos humanos cadavéricos. As soluções-corantes à base de Luteína e Zeaxantina poderão ser uma alternativa segura e eficaz em cirurgia de catarata e vitreorretiniana.

Point-of-care applications rely on biomedical sensors to enable rapid detection with high sensitivity and selectivity. Despite advances in sensor development, there are challenges in cancer diagnostics. Detection of biomarkers, cell receptors, circulating tumor cells, gene identification, and fluorescent tagging are time-consuming due to the sample preparation and response time involved. Here, we present a novel approach to target the enhanced metabolism in breast cancers for rapid detection using fluorescent imaging. Fluorescent analogs of fructose target the fructose-specific transporter GLUT5 in breast cancers and have limited to no response from normal cells. These analogs demonstrate a marked difference in adenocarcinoma and premalignant cells leading to a novel detection approach. The vastly different uptake kinetics of the analogs yields two unique signatures for each cell type. We used normal breast cells MCF10A, adenocarcinoma cells MCF7, and premalignant cells MCF10AneoT, with hepatocellular carcinoma cells HepG2 as the negative control. Our data indicated that MCF10AneoT and MCF7 cells had an observable difference in response to only one of the analogs. The response, observed as fluorescence intensity, leads to a two-point assessment of the cells in any sample. Since the treatment time is 10 min, there is potential for use in rapid on-site high-throughput diagnostics.
