Point‐of‐Care Platform for Rapid Multiplexed Detection of SARS‐CoV‐2 Variants and Respiratory Pathogens

Abstract: The rise of highly transmissible SARS‐CoV‐2 variants brings new challenges and concerns with vaccine efficacy, diagnostic sensitivity, and public health responses to end the pandemic. Widespread detection of variants is critical to inform policy decisions to mitigate further spread, and postpandemic multiplexed screening of respiratory viruses will be necessary to properly manage patients presenting with similar respiratory symptoms. In this work, a portable, magnetofluidic cartridge platform for automated pol… Show more

“…Although PCR amplification may require bulky and expensive lab equipment to perform the temperature fluctuation in the expansion process, as well as intense, trained human labor, Trick et al were able to overcome these limitations by integrating a magnetofluidic cartridge into an automated and portable PCR testing platform [ 120 ]. This tool utilized inexpensive components (e.g., 3D printing, laser cutting, and programed microcontroller) that combined the sample preparation steps and PCR to provide results in less than 30 min with minimal training required.…”

“…This tool utilized inexpensive components (e.g., 3D printing, laser cutting, and programed microcontroller) that combined the sample preparation steps and PCR to provide results in less than 30 min with minimal training required. The multiplexing is achieved by two fluorescence probes to identify B.1.1.7/B.1.351 variants and SARS-CoV-2/Influenza with a LOD of 12.5 copies/µL [ 120 ].…”

“…To date, researchers reported several POC devices for detection of SARS-CoV-2 ( Soares et al, 2021 ; Torezin Mendonça et al, 2022 ; Trick et al, 2022 ). These POC devices demonstrated a simple design and fast detection.…”

“…Owing to their high sensitivity and turnaround of 1-2 hours, NAATs are widely applied in medical diagnostics such as for HIV, tuberculosis, and SARS-CoV-2. [1][2][3] Polymerase chain reaction (PCR) and reverse transcription PCR are two of the most widely used techniques for detecting target DNAs or RNAs. Both techniques require a thermocycling process, which involves controlling the temperature of the reaction vessel to three distinct temperatures.…”
