Background and Purpose Non‐alcoholic fatty liver disease (NAFLD) is considered to be one of the most common chronic liver diseases across worldwide. Astaxanthin (Ax) is a carotenoid, and beneficial effects of astaxanthin, including anti‐oxidative, anti‐inflammatory, and anti‐tumour activity, have been identified. The present study aimed to elucidate the protective effect of astaxanthin against NAFLD and its underlying mechanism. Experimental Approach Mice were fed either a high fat or chow diet, with or without astaxanthin, for up to 12 weeks. L02 cells were treated with free fatty acids combined with different doses of astaxanthin for 48 h. Histopathology, expression of lipid metabolism, inflammation, apoptosis, and fibrosis‐related gene expression were assessed. And the function of mitochondria was also evaluated. Key Results The results indicated that astaxanthin attenuated HFD‐ and FFA‐induced lipid accumulation and its associated oxidative stress, cell apoptosis, inflammation, and fibrosis both in vivo and in vitro. Astaxanthin up‐regulated FGF21 and PGC‐1α expression in damaged hepatocytes, which suggested an unrecognized mechanism of astaxanthin on ameliorating NAFLD. Conclusion and Implications Astaxanthin attenuated hepatocyte damage and mitochondrial dysfunction in NAFLD by up‐regulating FGF21/PGC‐1α pathway. Our results suggest that astaxanthin may become a promising drug to treat or relieve NAFLD.

Eating habits and lifestyle directly impact general health. Consumption of fat- and sugar-rich foods and alcohol increase the risk of developing fatty liver disease. The prevalence of fatty liver disease is markedly critical, and its pathogenesis and progression are complicated. Chinese herbal medicine has been used to treat and prevent human diseases through-out history, and is a rich source of biologically active substances with unique curative properties. More recently, Chinese herbs and their extracts have been identified as a novel source of potential therapeutic agents in the prevention and treatment of fatty liver disease. Beneficial effects of these herbal medicines mean that they can be classified as novel candidates for the treatment and prevention of both alcoholic fatty liver disease (AFLD) and non-alcoholic fatty liver disease (NAFLD), in place of conventional allopathic treatments. In this review, we explore the current literature related to herbal medicines used for the treatment of or protection against fatty liver diseases and describe their mechanisms of action.

Background and aim Chrysin is a flavonoid found in plant extracts from Passiflora species, honey and propolis. It has demonstrated anti-adipogenic activity in vitro but there are no studies substantiating the anti-obesity activity of chrysin in vivo . Experimental procedure The pancreatic lipase (PL) inhibitory potential of chrysin was determined by preliminary in silico screening and further confirmed by in vitro PL inhibitory assay and oral fat tolerance test (OFTT). The effect of chrysin on acute feed intake and sucrose preference test was determined in normal rats. Obesity was induced by feeding of high fructose diet (HFD) to the rats. The rats were divided into six groups: normal control, HFD control, orlistat and three doses of chrysin (25, 50 and 100 mg/kg body weight). Body weight, body mass index (BMI), abdominal circumference/thoracic circumference (AC/TC) ratio, calorie intake, adiposity index, fecal cholesterol, locomotor activity and histopathology of the adipose tissue of the rats were evaluated. Results Chrysin showed good affinity to PL with competitive type of inhibition. It significantly reduced serum triglycerides in OFTT. Chrysin also significantly reduced acute feed intake and sucrose preference in rats. Chrysin significantly decreased the body weight, BMI, AC/TC ratio, adiposity index, calorie intake while it significantly increased the fecal cholesterol and locomotor activity of the rats. Chrysin was found to reduce the size of the adipocytes when compared to the HFD control group. Conclusion Thus, chrysin exerted anti-obesity effect by inhibiting PL, reducing sucrose preference, reducing calorie intake and increasing the locomotor activity of rats.
