PLP-dependent H2S biogenesis

Abstract: The role of endogenously produced H2S in mediating varied physiological effects in mammals has spurred enormous recent interest in understanding its biology and in exploiting its pharmacological potential. In these early days in the field of H2S signaling, large gaps exist in our understanding of its biological targets, its mechanisms of action and the regulation of its biogenesis and its clearance. Two branches within the sulfur metabolic pathway contribute to H2S production: (i) the reverse transsulfuration … Show more

“…In addition, homocysteine and cysteine were reported to be substrates for H 2 S production by the trans-sulfuration enzymes in mammals (39). Therefore, a possible mechanism of toxicity could be the conversion of SeHCys and/or SeCys to H 2 Se by cystathionine ␥-lyase (Cys3p).…”

“…Maintaining healthy levels of H 2 S in mammalian cells requires tight control of its biosynthesis and its catabolism (8,9). H 2 S is produced endogenously by two enzymes in the transsulfuration pathway, cystathionine β-synthase and γ-cystathionase as well as by mercaptopyruvate sulfurtransferase (MST) (10)(11)(12)(13). H 2 S is oxidized via the sulfide oxidation pathway to generate the end-products thiosulfate and sulfate via the concerted action of the mitochondrial enzymes sulfide quinone oxidoreductase, persulfide dioxygenase, rhodanese and sulfite oxidase (14)(15)(16).…”

“…For detailed information on the enzymology, tissue expression levels, and transcriptional and post-transcriptional regulation of CBS, we refer the reader to multiple specialized reviews (7,51,76,77,107). CBS (EC 4.2.1.22) is traditionally introduced as the first enzyme of the transsulfuration pathway.…”
