Platelet viability following storage for 5 days

Abstract: Regional blood centers frequently need to hold units of whole blood at 20 to 24 degrees C for several hours after phlebotomy so that sufficient platelet concentrates can be prepared to meet the increasing need. We have evaluated the in vivo viability and in vitro properties of platelets that were prepared from whole blood drawn into citrate-phosphate-dextrose-adenine (CPDA-1) either immediately after phlebotomy or after an 8-hour hold at 20 to 24 degrees C. Platelet concentrates were stored for 5 days at 20 to… Show more

“…These recovery and survival studies have been initiated. It has also been shown that red cell and platelet preparations from standard CPD.Al dona tions may be prepared 8-15 h after donation [43][44][45]. It will be of interest to determine if this is also possible for half-strength citrate donations and if so whether the improved stability of Vili : C already demonstrated for blood (table II) and plasma ( fig.…”

“…Other workers have already demonstrated that red cell and platelet preparations may be prepared up to 8 [6,[9][10][11][12][13][14][15] or 15 h [ 16] after collection of blood in CPD anti coagulants. The in vitro data presented here indicate that red cell and platelet preparations of good quality may be prepared 18 h after collection in 0.5 CPD.A2.…”

“…In human medicine, PCs are usually used for platelet administration and PCs are transfused within 72 hr after blood drawing [6,15,20]. Even the lyophilized dry technique of platelets was investigated to extend storage time [19].…”

“…Changes of platelet aggregability were assessed in this study also. Because platelet aggregation in CPDA-1 treated PRP is much weaker than that in 0.38% sodium citrate treated PRP [15], CaCl 2 was added to counteract the excess citrate and therefore enhance aggregation. Groh et al [10] also performed the re-calcification technique for platelet aggregometry using PRP treated with acid citrate dextrose, also a blood cell preservative but not an ideal anticoagulant for platelet aggregometry.…”
