Platelet-rich plasma combined with agarose as a bioactive scaffold to enhance cartilage repair: An in vitro study

Abstract: Platelet-rich plasma showed a positive effect on chondrocytes proliferation, differentiation and integration between native cartilage and engineered tissue when combined with agarose gel. Our findings suggest that platelet-rich plasma-agarose gel scaffold is a promising bioactive scaffold for future cartilage tissue engineering and future clinical works.

“…Platelets are a known source of several GF with beneficial effects in tissue healing 4 , including transforming growth factor-beta (TGF-β), insulin-like GF I (IGF-I), platelet-derived GF (PDGF), vascular endothelial GF (VEGF), fibroblast GF (FGF), and epidermal GF (EGF). This knowledge stimulated the development of a platelet concentrate with the purpose of increasing levels of local GF delivery at the injury site and improving the tissue repair process 5 .…”

“…The porous scaffold may also contain biological factors that promote cell proliferation and differentiation. 21,22 To improve the biological effects, numerous attempts have been set up to promote cell adhesion and ECM secretion of 3D PCL scaffolds by combining them with degradable bioactive materials (BMSC-derived natural ECM, 23,24 platelet-rich plasma, [26][27][28] and collagen 29 ). Our previous study testified that MC is able to regenerate cartilage by using biodegradable BMSC-derived ECM (BMSC-dECM) scaffolds.…”

“…7,8 However, the last and probably the most important mechanism of action that explains the well-documented pain reduction, which is also the most prominent and disabling symptom of knee OA, seems to be the anti-inflammatory and antinociceptive role (by regulation of nuclear factor-kappa B and cyclooxygenase-2).…”
