Platelet extracellular vesicles induce a pro‐inflammatory smooth muscle cell phenotype

Abstract: Extracellular vesicles (EVs) are mediators of cell communication during health and disease, and abundantly released by platelets upon activation or during ageing. Platelet EVs exert modulatory effects on immune and vascular cells. Platelet EVs may modulate the function of vascular smooth muscle cells (SMC). Platelet EVs were isolated from platelet-rich plasma and incubated with SMC in order to assess binding, proliferation, migration and pro-inflammatory phenotype of the cells. Platelet EVs firmly bound to res… Show more

“…Intravenous injection of endothelial-derived EVs loaded with microRNA-126 prevents SMC proliferation, migration, and neointima formation in a mouse model of vascular injury. Similarly, platelet-derived EVs can induce a pro-inflammatory remodeling phenotype in SMCs, including increased migration and proliferation [9]. Systemic pressures (e.g., introduction of pathological agents) often first effect blood cells and the endothelial cells that line the blood vessel.…”

“…Samples were analyzed under constant flow conditions (flow rate = 50) at 25 °C. Fifteen 60 s videos were captured with a camera level of 16 and detection threshold of 5 as before . NTA analysis revealed a shift in the distribution of small (0–200 nm) vesicles in pregnancy.…”

“…Receiver operator characteristic (ROC), AUC, specificity, and sensitivity were calculated using the ROCR package . Particle size distribution was determined by NTA using a Malvern NanoSight NS300 (Malvern Technologies, Malvern, UK) with a high‐sensitivity sCMOS camera and a 488 nm laser as before . Data was analyzed using NTA 3.1.54 software.…”
