Plasma MicroRNAs Relate to Atrial Fibrillation Recurrence after Catheter Ablation: Longitudinal Findings from the MiRhythm Study

Abstract: Introduction: Genetic and transcriptomic factors play important roles as mediators of new-onset and recurrent atrial fibrillation (AF). MicroRNAs (miRNAs) regulate expression of gene networks involved in key aspects of atrial remodeling. Associations between circulating miRNAs and AF recurrence are unknown. We tested the hypothesis that cardiac miRNAs associated with electrical and structural remodeling predict recurrent AF rhythm in post-ablation patients.

“…The circulating miR-21 level is also associated with AF ablation results in patients with persistent AF undergoing ablation (and this level correlates with left atrial low voltage areas) 8 . Other studies have shown higher levels of miRNAs 125a, 10b, 601, 30a-3p, and 199b in patients with 12-month AF recurrences after catheter ablation than in patients without AF recurrences 9 .…”

“…miRNAs regulate the molecular processes that trigger the structural and electrophysiological remodeling key to the development of AF (69, 21). They are associated with incident and recurrent AF, and their dynamic nature after catheter ablation suggests that they are related to the pathogenesis of AF (5, 15, 21). In this work, we explore the relationship between iLAEAT and plasma miRNA and hypothesize their potential role in creating a substrate for AF.…”

“…As part of the miRhythm study examining links between circulating miRNAs and AF, 584 participants were recruited at the University of Massachusetts Medical Center (UMMC) between April 2011 to July 2017 (15). Study protocol was approved by the University of Massachusetts Institutional Review Board (IRB #14875).…”

“…TBX5 is a target gene of both miR-10a and miR-10b [84]. Expression of miR-10b was found to be upregulated in LA biopsies from patients with chronic AF and valvular heart disease [27] and miR-10b plasma levels are decreased in AF patients after catheter ablation [85]. In the porcine model, shortlasting atrial tachyarrhythmia was associated with significant upregulation of both miR-10a and miR-10b coupled with decreased TBX5 expression in the LA of paced animals that may be permissive for AF development [17].…”
