Plant SUMO E3 Ligases: Function, Structural Organization, and Connection With DNA

Abstract: Protein modification by the small ubiquitin-like modifier (SUMO) plays an important role in multiple plant processes, including growth, development, and the response to abiotic stresses. Mechanistically, SUMOylation is a sequential multi-enzymatic process where SUMO E3 ligases accelerate SUMO conjugation while also influencing target identity and interactions. This review explores the biological functions of plant SUMO E3 ligases [SAP AND MIZ1 DOMAIN-CONTAINING LIGASE (SIZs), METHYL METHANESULFONATE-SENSITIVIT… Show more

“…SUMOylation protects conjugated protein from degradation by blocking the lysine residues that can be ubiquitinated ( Roy and Sadanandom, 2021 ). Thus, SUMOylation of ICE1 by SAP AND MIZ1 DOMAIN-CONTAINING LIGASE 1 (SIZ1; Jmii and Cappadocia, 2021 ) protects ICE1 from degradation through the 26S proteosomic degradation pathway. Expression of CBF/DREB1 and the downstream COR genes was increased while the expression of MYB15 was suppressed due to SIZ1 conjugation of ICE1, enhancing the tolerance to low temperature ( Miura et al, 2007 ).…”

“…Abiotic stress is essentially any environmental condition that negatively affects the growth and general development of a plant [ 3 ]. This may range from mechanical disturbances, such as strong winds, to rapid changes in light conditions on a cloudy day, but generally it is mostly connected with severe stress conditions, such as soil salinity, heat waves, and drought stress [ 3 , 4 , 5 ]. The sessile lifestyle of plants makes it impossible for them to avoid these stressors by simply changing their location; hence, they need to have strategies in place to rapidly respond to changes in their environment in order to survive and sustain their growth.…”

“…E3 ubiquitin ligases, the most abundant eukaryotic proteins (Mazzucotelli et al 2006 ; Liyuan et al 2013; Michael et al 2015 ), catalyze the covalent attachment of ubiquitin to substrates with the help of E1 (ubiquitin-activating enzyme) and E2 (ubiquitin-conjugating enzyme) proteins (Buetow and Huang 2016 ). A wide variety of ubiquitination processes participate in numerous biological pathways (Jmii and Cappadocia 2021 ; Wu et al 2021 ; Liu et al 2021 ; Uchida and Kitagawa 2016 ), and the predominant function, mediated by Lys48-linked ubiquitination, is to target proteins to the proteasome for degradation (Hiranyakorn et al 2020 ). Other types of linkages, e.g., Lys63-linkages, signal substrates for non-degradative functions, such as protein assembly or modification (Chen and Sun 2009 ) (Swatek and Komander 2016 ); or chromatin modelling or repair (Nakazawa et al 2020 ).…”
